Effect of α-synuclein on the promoter activity of tyrosine hydroxylase gene

Abstract: Objective To approach the associated mechanism by which α-synuclein (α-Syn) might regulate the metabolism of dopamine. Methods A DNA fragment, located at -495 to +25 of the human tyrosine hydroxylase (TH) gene, was amplified by PCR and inserted into the pGL 3 -Basic luciferase reporter vector. The recombinant plasmid pGL 3 -THprom was transfected into a dopaminergic cell line MES23.5 or a α-Syn over-expressed MES23.5 (named MES23.5/hα-Syn + ). The promoter activity was detected by the Dual Luciferase Assay Sys… Show more

“…The physiological role of nuclear a-Syn remains elusive, and only few data have demonstrated that a-Syn affects gene expression and regulates TH expression in both cultured cells and brain of aged monkeys (Chu and Kordower, 2007;Gao et al, 2007;Wu et al, 2009). …”

“…Whereas the physiological function of α-synuclein remains to be fully characterized [63-65]. Several studies have established that interaction of α-synuclein and TH is of functional relevance to dopaminergic neurotransmission and the pathophysiology of PD [49, 66, 67]. Indeed, α-synuclein gene mutations and PD pathogenesis are well documented [49].…”
