Effect on tumorigenicity and metastasis of transfection of a diploid benign rat mammary epithelial cell line with DNA corresponding to the mRNA for basic fibroblast growth factor

Abstract: To examine the potential role of fibroblast growth factors (FGF) in tumorigenesis and metastasis, plasmid constructs containing the human basic FGF (bFGF) gene, with or without fusion to a secretory signal peptide (IgbFGF), were transfected into the diploid rat mammary epithelial cell line Rama 37. All transfectants possessed multiple copies of the transfected cDNA, which was expressed as the corresponding mRNA and the protein. The amount of bFGF protein was usually greater than the bFGF growth-stimulatory act… Show more

“…Thus, when FGF-2 mRNA levels were analysed in a cohort of breast cancer patients, elevated expression of FGF-2 mRNA correlated with a good prognostic outcome, the opposite of the result expected from the naïve perspective that "FGF2 = uncontrolled growth + angiogenesis = cancer" (Anandappa et al 1994). Similarly, in a syngeneic rat model of breast cancer, overexpression of FGF2 failed to produce any metastases (Davies et al 1996). Given the difficulty in publishing negative results, there is likely a very large body of work that demonstrates the absence of a direct association between the expression of FGF ligands and cancer.…”

“…Isolation and characterization of the activity determined that FGF8 contributed to some of the growth effects. In humans, it is present in increased concentrations in breast cancer cells (Zammit et al, 2002), and its 22-protein family has a wide range of effects from wound healing repair (Clarke et al, 1993;Cuevas, 1998) to tumeriogenesis (Davies et al, 1996). In cattle, there are no reports linking FGF8 to variations in carcass quality.…”
