Yellow rust caused by Puccinia striiformis f. sp. tritici is the most devastating fungal disease of wheat, especially in the Central and West Asia and North Africa (CWANA) region. Most of the released cultivars in CWANA have become susceptible to the new virulence Yr27. Growing durable resistance cultivars is the most economical control measure. A field study was conducted to evaluate 500 bread wheat landraces along with the susceptible control 'Morocco' using artificial inoculation under field conditions at Tel Hadia, Syria during 2010/11 and 2011/12 growing seasons. The most prevailing and spreading yellow rust virulence (70E214) was used for artificial inoculation. Disease scoring started when disease severity was more than 50% on the leaves of cv. Morocco and continued for four readings at 7 days intervals. Slow rusting resistance was assessed based on development of the disease over time using the area under disease progress curve (AUDPC), coefficient of infection (CI), final rust severity (FRS), infection rate "r" and relative resistance index (RRI). None of the landraces showed immune reaction and 10% showed the lowest values for all parameters, suggesting that major genes control resistance in these landraces. Approximately 65% of the landraces were marked as having different levels of slow rusting and 25% were highly susceptible. Cluster analysis based on partial resistance parameters of the landraces along with cv. Morocco revealed two major clusters: susceptible and low level of slow rusting were grouped in the first cluster; resistant, high level and moderate level of partial resistance were grouped in the second cluster. These groups were also confirmed using principal coordinate analysis. By comparing the results of RRI and others parameters, it appeared that landraces with very low values exhibited high RRI value of 9, while those that showed high, moderate, and low levels of slow rusting, had RRI ranges of 8-9, 7-8 and 5-7, respectively. Landraces with maximum values from each parameter showed very low RRI values of less than 5. The results also suggest that AUDPC, CI, and FRS are the more appropriate parameters for assessing slow rusting than infection rate. Correlation coefficient and principle component analysis revealed a high and positive correlation between these parameters.

Wheat stripe rust, which is caused by the basidiomycetes fungal pathogen Puccinia striiformis f.sp. tritici (Pst), is one of the most destructive diseases of wheat in Türkiye and worldwide. In this study, a total of 99 bread wheat varieties were assessed at the adult plant stage under natural infection conditions during the 2021–2022 and 2022–2023 growing seasons. Additionally, specific molecular markers were used to genotype the stripe rust resistance genes Yr26, Yr32, Yr36, Yr44 and YrSP. Disease severity was determined using the modified Cobb scale and infection reactions of each bread wheat varieties were evaluated, and also coefficient of infection value was calculated based on those parameters. To determine whether the wheat varieties had resistance gene(s), specific molecular markers associated with the resistance genes Yr26, Yr32, Yr36, Yr44 and YrSP were used. In the field trials, the majority of the tested varieties showed moderately susceptible and susceptible reactions in both years. According to the molecular results, they were positive for Yr32 in three varieties, Yr36 in 25 varieties, YrSP in two varieties and none of them had Yr26. Furthermore, some varieties carried combinations of these genes, such as Yr32 + Yr36, Yr32 + Yr44, Yr36 + Yr44, Yr36 + YrSP, Yr32 + Yr36 + Yr44 and Yr32 + Yr36 + Yr44 + YrSP. On the other hand, it was not determined these resistance genes in 16 varieties. In the literature, these resistance genes were first identified in Turkish bread wheat varieties. In addition, these findings are valuable for the development of wheat cultivars with effective and more durable resistance to stripe rust in Türkiye.

Stripe (yellow) rust, caused by Puccinia striiformis f.sp. tritici (Pst), is a fungal disease that presents one of the most serious threats to the wheat crops, causing severe yield losses worldwide, including Kazakhstan. The objectives of this study were to: (1) evaluate a winter wheat collection for stripe rust resistance during an adult plant growth stage, (2) identify the presence of selected Yr genes using linked molecular markers in wheat germplasm, (3) identify potentially useful resistant wheat genotypes among leading cultivars and advanced breeding lines. This study evaluated 70 winter wheat genotypes for stripe rust resistance. According to the field reactions, 42 entries (60%) had R or MR reactions including 27 breeding lines (38.6%) and 15 (21.4%) cultivars. Twenty-eight breeding lines/cultivars (40.0%) were susceptible in both years. According to the average coefficient of infection value (ACI) six genotypes were regarded as possessing high level of adult plant resistance. Cultivars/lines carrying Yr10 alone or in combination with other Yr resistance genes provided resistance to stripe rust. Eleven breeding lines showed <5% disease severity in both years. Linked marker analysis revealed the presence of several gene and gene complexes (Yr5, Yr10, Yr15, Yr17/Lr37/Sr38 and Yr18/Lr34). Among a collection of 70 winter wheat breeding lines and cultivars produced in Kazakhstan three stripe rust resistance genes (Yr10, Yr5 and Yr15) demonstrated high frequency occurrence (31.4%, 14.0% and 7.0%, respectively). The most abundant was gene Yr10 identified in 22 genotypes. It was followed by the Yr5 gene, which conferred resistance in 14 lines (20%) and Yr18 gene-11 lines (15.7%). Yr15 was identified in 7 genotypes. Yr17/Lr37/Sr38 gene complex was found in 2 entries. Among 70 evaluated germplasm sources, 42 disease resistant entries are potentially useful resistant wheat genotypes. These carriers of different Yr genes can be used directly in breeding programs to improve stripe rust resistance of winter wheat. Marker-assisted selection can be efficiently applied to develop wheat cultivars with effective gene combinations that would directly assist in developing durable resistance in Kazakhstan.
