Effective Delivery of siRNA-Loaded Nanoparticles for Overcoming Oxaliplatin Resistance in Colorectal Cancer

Abstract: Chemotherapy resistance represents a formidable obstacle in advanced or metastatic colorectal cancer (CRC) patients. It is reported that ATPase copper transporting alpha (ATP7A) plays an important role in chemotherapy resistance in CRC. Here, we identified ATP7A as a potentially key gene of OXA resistance in CRC. The patients with higher expression of ATP7A tended to have platinum drug resistance. While the lower expression of ATP7A by siRNA knockdown resulted in enhancement of OXA sensitivity and increased OX… Show more

“…We performed qRT-PCR analysis as previously described [ 48 , 49 , 50 ]. Briefly, total RNA was extracted by Total RNA Isolation Reagent (BS258A, Biosharp, Shanghai, China) and reverse transcribed into complementary DNA (cDNA) by the First-strand cDNA Synthesis Kit (K1072, Apexbio, Huston, TX, USA).…”

“…ATP7A is one of the key regulators of the intracellular level of platinum-based drug such as Oxaliplatin (OXA) [ 105 ], commonly used to treat colon cancer patients. Based on this knowledge, Zhou et al [ 99 ] delivered to the colon cancer cells HCT116 and LOVO a siRNA (siATP7A) by means of a lipid based delivery system. Besides reaching good down-regulation in the expression of the target gene, the authors could also show that cells treated by siATP7A displayed increased sensibility to OXA.…”

“…Strategy of co-delivering chemotherapeutics and siRNA targeting chemoresistance-related machinery has been proposed and studied widely in treating malignant cancers such as colorectal cancer. 22 , 23 Polymer nanoparticles 24 and inorganic nanoparticles 25 have been used as nanoplatform for co-delivering siRNA and drugs. They discovered that co-delivery strategy not only enhances drug delivery through penetration into tumor tissues but also antagonizes chemoresistance by modulating chemoresistance-associated genes.…”
