Reproductive activities of salmonids are synchronized by changes in photoperiod, which control the endocrine system via the brain-pituitary-gonadal axis. Gonadotropin-releasing hormone (GnRH) in the brain regulates synthesis and release of the pituitary gonadotropins (GTHs; FSH and LH). FSH and LH in turn stimulate the production of sex steroids for oocyte growth and maturation-Inducing steroid hormones for oocyte maturation and ovulation, respectively, in female salmonids. To clarify effects of long-term photoperiod manipulations on the reproductive activity of salmonids from early recrudescence to ovulation, we Investigated the gene expression profiles of GnRH, GTHs, and vitellogenin (VTG), and plasma sex steroids in female rainbow trout (Oncorhynchus mykiss). In addition, the percentages of eyed embryos and hatched alevins were examined together with the number of ovulated eggs to evaluate the effects of photoperiod regimes on egg quality. During late summer, the mRNA levels of GnRHs, GTHalpha, and LHbeta, and the plasma level of a maturational steroid (17alpha,20beta-dihydroxy-4-pregnen-3-one; 17,20beta-P) were significantly elevated by a gradually shortened photoperiod under constant temperature, in accordance with accelerated sexual maturation. The percentages of eyed embryos and hatched alevins from fish ovulated in August were comparable to those of control fish observed in December. These results clearly indicate that syntheses of GnRHs, LH, VTG, and 17,20beta-P are effectively accelerated by a programmed long-short photoperiod regime in early recrudescent female rainbow trout, without a marked deterioration in egg quality.

ABSTRACT:The effect of single and double injections of D-Tle 6 ,Pro 9 ,NEt-mGnRHa (Supergestran ® ) on advancement and induction of ovulation in Arctic grayling was assessed. Sexually mature wild Arctic grayling females (most 2-4 years old) were caught in the Yenisey River at the beginning of May 2010. After a 4-day acclimatization, the females were randomly divided into four groups and intramuscularly injected as follows: group A, control group, treated with physiological saline only; group B, treated with a single injection (SI) of Supergestran ® at 25 µg/kg body weight (BW); group C, injected twice (DI) with 25 µg/kg BW 3 days apart; group D, injected twice with 10 µg/kg BW 3 days apart. After stripping, the pseudo-gonadosomatic index was calculated, and an eggs sample from each female was fertilized. Only fish in the groups treated with DI protocols ovulated. No differences between the two groups were found in the timing of ovulation, ovulation rate, or mean time to ovulation. No females in either group A or B ovulated, since the experiment had to be prematurely terminated due to technical problems at the field hatchery. The DI of 10 µg/kg proved sufficient to induce and advance ovulation in Arctic grayling. Hormone treatments seem to be a promising tool to obtain viable eggs of Arctic grayling in a short time window and thereby to ensure satisfactory numbers of fry for restocking programs.

Various induction protocols were compared with the aim to reach ovulation in northern pike (Esox lucius L.). The effectiveness of salmon D-Arg 6 Pro 9 NEt-GnRHa at two doses in combination with various additives were tested: 50 and 100 µg kg -1 (groups GnRHa 1 and 2), GnRHa at 50 and 100 µg kg -1 in combination with 8 mg kg -1 metoclopramide (groups 3 and 4), GnRHa at 50 and 100 µg kg -1 emulsified in Freund´s incomplete adjuvant (FIA) (groups 5 and 6), and GnRHa at 50 and 100 µg kg-1 emulsified with FIA 8 mg kg -1 and metoclopramide (groups GnRHa 7 and 8). These treatments were compared to indoor controlled conditions (Group ICC, injected with saline solution, without hormonal treatment) and two traditional treatments: 3 mg kg -1 carp pituitary (Group CP) and ambient outdoor conditions (Group AOC, injected with saline solution only, without hormonal treatment).All fish in Group CP and 70% of those in Group AOC ovulated. The latency periods for Group CP was 96±4 h and 264±58 h for Group AOC. One Group-3 female ovulated spontaneously 108 h post-injection, whereas none of the other GnRHa injected or Group ICC fish ovulated. The pseudogonadosomatic index was 19.3±5.9%; 18.5% and 17.8±7.5% in the AOC, Group 3 and CP, respectively. The fertilization rate reached 88.6±4.5%; 85.5±12.0% and 66.0±13.7% in AOC, Group 3 and CP, respectively. The hatching rate was 68.6±9.9%; 65.5±7.5% and 54.4±8.0% in AOC, Group 3 and CP, respectively.The ovarian fluid pH was significantly higher (8.27±0.03) in AOC compared to CP group (8.11±0.02) (P>0.05). Ovarian fluid of the spontaneously ovulated Group-3 fish had a pH of 8.35. There were no significant differences in egg size and weight among successfully reproduced groups -AOC and CP (P>0.05).
