Effects of a lipase inhibitor (Orlistat) on cholecystokinin and appetite in response to a high-fat meal

Abstract: OBJECTIVE:To examine the short-term effects of a lipase inhibitor (Orlistat) on physiological and behavioural measures of appetite in response to a high-fat meal. DESIGN: Randomised, single blind, placebo-controlled, crossover trial. SUBJECTS: A total of 19 healthy nonobese male subjects. PROCEDURES: After an overnight fast, subjects ingested a test meal of 2940 kJ (60% fat, 30% CHO, 10% protein) with Orlistat (120 mg) or a placebo, separated by 2 weeks. Appetite, as assessed by a standard line scale, and plas… Show more

“…It has been shown that fat induces satiety as long as it stays in the intestine (Beglinger and Degen, 2004;Goedecke et al, 2003). The satiety signals are released from the gastrointestinal tract triggered by the entry of food products.…”

“…The use of lipase inhibitors in pharmaceuticals targeting obesity is well established, Orlistat being present at the market today. A known side effect of this drug and similar lipase inhibitors is steatorrhea due to impaired lipid digestion 4 . An alternative to a pharmaceutical approach to lipase inhibition is to use food-based components to prolong lipid digestion and increase satiety, yet without causing negative side effects that reduce patient compliance.…”
