“…As a result of the work, cells of the HEK293T line, which are capable of converting cholesterol and its analogues to pregnenolone, were obtained and characterized. It was found that the activities of the obtained cells, which were transfected with pcDNA 3.1/pCoxIV‐CHL‐GFP as well as pcDNA 3.1/pCoxIV‐P450‐2A‐GFP plasmid, were comparable to the activity of natural steroidogenic cells–0.0012 to 0.0025 ng of pregnenolone/cell/24 hours for transfected HEK293T in This study versus 0.00008 ng of pregnenolone/cell/hour for rat granulosa cells; 0.0019 ng of pregnenolone/cell/h, 0.00005 ng of pregnenolone/cell/hour and 0.000044 ng of pregnenolone/cell/hour for rat adrenal cortex cells (25‐OH‐cholesterol as substrate of P450scc and trilostane as inhibitor of 3β‐HSD were used in all experiments).
