Effects of Acute Intravenous Infusion of Apelin on Left Ventricular Function in Dogs With Advanced Heart Failure

Abstract: Background Apelin-13 (APLN) through apelin receptor (APJ) exerts peripheral vasodilatory and potent positive inotropic effects. We examined the effects of exogenous intravenous infusion of APLN on left ventricular (LV) systolic function in dogs with heart failure (HF, LV ejection fraction, EF~30%). Methods and Results Studies were performed in 7 dogs with microembolization-induced HF. Each dog received an intravenous infusion of low dose and high dose APLN followed by washout period. LV end-diastolic volume … Show more

“…Apelin production falls when HF becomes chronic or more severe [76]. An example of this fall is the reduction of the myocardial level of apelin observed in the dog with an advanced HF induced with microembolization of a coronary artery [78]. The biphasic changes in apelin release are in line with the general opinion that in the early stages of HF, factors activating the force of contraction, as e.g.…”

“…4 In a recent study, exogenous administration of apelin improved of left ventricular systolic function in dogs with advanced heart failure. 23 In 2008, Azizi et al investigated the effect of hypertonic saline infusion or water loading on osmolality, apelin and arginine vasopressin (AVP). Increasing plasma osmolality was accompanied by a parallel, linear increase in plasma AVP concentration and by a decrease in plasma apelin concentration.…”

“…These dynamics are considered to be an attempt to compensate and increase the contractile force of the failing heart [22], but these effects are progressively altered in the course of heart failure. These assumptions were made after observing the effects of experimental iv administration of apelin to animals and humans with heart failure: it acts as a potent inotrope and increases the contractility of the heart, the ejection fraction and the cardiac output and lowers the left ventricle diastolic pressure .…”
