Effects of an isogenic Zn‐metalloprotease‐deficien mutant of Legionella pneumophila in a guinea‐pig pneumonia model

Abstract: To determine the effects, if any, of the Zn-metalloprotease on virulence of Legionella pneumophila infection, an isogenic mutant deficient in protease (encoded by the proA gene) was tested in an Acanthamoeba cell model, in guinea-pig macrophages, and in a guinea-pig pneumonia model. The cloned proA gene was completely inactivated by insertion of a kanamycin-resistance cassette into the protease gene of L. pneumophila AA100. This mutated gene was then introduced into the L. pneumophila chromosome by allelic exc… Show more

“…It degrades host extracellular matrix and immunity proteins, supports Hartmanella vermiformis amoebae and guinea pig infection, and is indeed found in the L. pneumophila phagosome (62)(63)(64). Here we showed that the enzyme also regulates lipolytic activities of the pathogen that might contribute to the observed loss of hemolytic activities in a L. pneumophila proA knock-out mutant (65,66).…”

“…Given this apparent disparity from the rest of the d N /d S results, a more detailed examination was undertaken. The mature protein product of proA is a secreted metalloprotease which has been shown to have broad specificity and cytotoxic activity (Black et al, 1990;Moffat et al, 1994). In the SBT scheme, the fragment of proA that is used to define the allele (reference sequence M31884; positions 1134-1538) corresponds approximately to amino acids 139-276 of the mature protein.…”

“…The enzyme hydrolyzes a broad spectrum of protein substrates and confers hemolytic as well as cytolytic activities (47,53). Additionally, ProA has been shown to contribute to bacterial pathogenesis in a guinea pig model of pneumonia (38). Another hemolytic, but not cytotoxic, protein is the L. pneumophila legiolysin Lly, which is also responsible for color production and fluorescence of the bacterium (61).…”
