Effects of Anions on ATP-Induced [Ca2+]i Increase in NG108-15 Cells

Abstract: ABSTRACT-We investigated the effects of anions on different P2 receptors by measuring ATP-induced increase in intracellular Ca2+ concentration ([Ca 2+ ]i) in fura-2-loaded NG108-15 and PC12 cells. In NG108-15 cells, ATP at 100 mM and 1 mM induced a transient and a sustained [Ca 2+ ]i increase, respectively. The former, but not the latter, was inhibited by U-73122, indicating that the former was via the P2Y2 receptor and the latter via the P2X7 receptor. When external Cl -was replaced by other anions, th… Show more

“…In the present study, we have clearly demonstrated that the P2Y 2 receptor and G q/11 are present in the cholesterolrich lipid rafts that are required for effective signal transduction, and this signal transduction via G q/11 is essential for It has been shown that ATP interacts with P2X 7 and P2Y 2 receptors expressed in NG 108-15 cells (Watano et al, 2002). CD is reported to extract the cholesterol followed by disruption of the cholesterol-dependent lipid rafts on the plasma membrane in NG 108-15 cells.…”

“…Endothelial cells are known to possess both P2X and P2Y receptors (6 -9, 26). We attempted to separate the response mediated by P2X receptors from that by P2Y receptors by using the phospholipase C inhibitor U73122, as we described previously in other cell types (27,28). Although U73122 (2 μM) decreased the Ca ] i elevation in IFN-γ-treated cells remained even in the presence of U73122 (Fig.…”

“…ATP and UTP were shown to increase [Ca 2+ ] i in the murine neuroblastoma cell line NIE-115 [542], perhaps via P2Y 2 and/or P2Y 4 receptors. Later, P2X7 receptor mRNA and protein were shown to be present on NIE-115 cells, mediating apoptosis, perhaps via breakdown to adenosine [543], but more likely by direct activation [544]. P2U (i.e.…”
