Effects of culture conditions on mycelium biomass and intracellular cordycepin production ofCordyceps militaris in natural medium

Abstract: -Effects of culture conditions on the growth of mycelium and the production of intracellular cordycepin of Cordyceps militaris were investigated in this study. The natural medium which is a mixture of 53% brown rice paste, 6% beerwort and 42% soybean meal juice was used for the fermentation in shaking flasks. Fermentation temperature, pH and medium capacity, which had been proved to be significant factors, were optimized by Box-Behnken design. Results showed that dry mycelia weight (DMW) and cordycepin yield (… Show more

“…The optimal pH for cordycepin production by different strains of C. militaris has been reported to be in the range of 4-7 (Kang et al, 2014;Zhong et al, 2011). Other studies reported that C. militaris strain BCC2816 achieved the highest dry mycelial mass (19.1 g l -l ), and optimal cordycepin yield (1.8 mg l -l ) at 28 °C, pH 6.2 (Chunyan et al, 2009). In general, the effects of pH in static culture of strains AG-1, PSJ-1 in the present study were lower compared with other studies.…”

“…In previous work, the orthogonal design method [ 44 46 ], Box-Behnken design [ 34 , 47 ], and central composite design [ 31 ] were used to optimize culture conditions for cordycepin production by Cordyceps sp. These experimental designs have been successfully used to optimize medium for the mycelial growth and microbial metabolite production in liquid culture processes.…”

“…Many studies have reported that oxygen supply could evidently influence the formation and accumulation of bioactive metabolites in the submerged cultivation of medicinal fungus [24]. Xie et al [25] also reported that with the increasing of medium capacity dry mycelia weight decreased but cordycepin production enhanced, and obtained the highest cordycepin yield of 1.8 mg g -1 .…”
