Effects of diaspirin crosslinked hemoglobin infusion in treadmill-exercised swine

Abstract: The hemodynamic and metabolic effects of diaspirin crosslinked hemoglobin (DCLHb) were investigated using graded treadmill exercise in swine (n = 5/group). Swine received DCLHb (10% solution, 5 ml/kg) or oncotically-matched human serum albumin (HSA, 5ml/kg). Baseline metabolic and hemodynamic data were similar. In both groups exercise increased hemodynamic parameters. Exercise increased heart rate (HR) from 139 +/- 12 to 293 +/- 28 bpm with DCLHb and from 136 +/- 13 to 314 +/- 13 bpm with HSA. Exercise increas… Show more

“…In animals, Crago et al [3] studied five healthy, nonanaemic swine infused with 5 ml/kg of a 10% diaspirin crosslinked haemoglobin solution (DCLHb) during graded treadmill exercise, and reported a higher oxygen extraction in the DCLHb group accompanied by a decrease in cardiac output. This effect was evident during both exercise and the recovery periods, and was interpreted to be indicative of an enhanced capacity for oxygen delivery mediated by the HBOC solution.…”

“…Contrasting this, Crago et al [9] showed in five healthy, non blood-depleted swine that after infusion of 5 ml/kg of 10 % DCLHb-solution, oxygen delivery and recovery was improved during graded treadmill exercise. Oxygen debt was reduced due to higher oxygen delivery to the tissue.…”

“…In addition to the restoration of intravascular volume and oxygen, HBOC seem to have a higher oxygenation potency when compared to RBCs [75] because of their plasmatic oxygen transport and enhanced oxygen release from the HBOC themselves and from adjacent erythrocytes [76]. The increased oxygen extraction seems to be a characteristic essential of modern cell-free hemoglobin and has been proved in different animal models [75,77]. There is evidence from animal studies that the vasoconstrictive side effects associated with HBOC treatment, whether they are caused by NO scavenging [78], endothelin release [79] or modulation of alpha-adrenoreceptors [80], are more than compensated by the enhanced oxygen off-load through capillaries.…”
