Low-pathogenicity avian influenza H9N2 remains an endemic disease worldwide despite continuous vaccination, indicating the need for an improved vaccine strategy. Bacillus subtilis (B. subtilis), a gram-positive and endosporeforming bacterium, is a non-pathogenic species that has been used in probiotic formulations for both animals and humans. The objective of the present study was to elucidate the effect of B. subtilis spores as adjuvants in chickens administered inactivated avian influenza virus H9N2. Herein, the adjuvanticity of B. subtilis spores in chickens was demonstrated by enhancement of H9N2 virus-specific IgG responses. B. subtilis spores enhanced the proportion of B cells and the innate cell population in splenocytes from chickens administered both inactivated H9N2 and B. subtilis spores (Spore + H9N2). Furthermore, the H9N2 and spore administration induced significantly increased expression of the pro-inflammatory cytokines IL-1β and IL-6 compared to that in the H9N2 only group. Additionally, total splenocytes from chickens immunized with inactivated H9N2 in the presence or absence of B. subtilis spores were restimulated with inactivated H9N2. The subsequent results showed that the extent of antigen-specific CD4 + and CD8 + T cell proliferation was higher in the Spore + H9N2 group than in the group administered only H9N2. Taken together, these data demonstrate that B. subtilis spores, as adjuvants, enhance not only H9N2 virus-specific IgG but also CD4 + and CD8 + T cell responses, with an increase in pro-inflammatory cytokine production. This approach to vaccination with inactivated H9N2 together with a B. subtilis spore adjuvant in chickens produces a significant effect on antigenspecific antibody and T cell responses against avian influenza virus.

β-mannan is a nonstarch polysaccharide found in hulled and dehulled soybeans that can survive drying-toasting phase of processing soybeans and have antinutritive effects in poultry. β-mannanase is an active enzyme (endohydrolase) that can hydrolyze β-mannan to reduce its antinutritional effects. Two experiments were conducted to evaluate the effects of β-mannanase supplementation in low energy/protein diets on egg production, egg quality, and apparent ileal digestibility of the dry matter ( DM ), crude protein ( CP ), and amino acids in 21-week-old Single Comb White Leghorn hens (Hy-Line W-36). A total of 192 hens (8 replicates of 6 hens per treatment) for a production study (Exp. 1) and a total of 64 hens (8 replicates of 2 hens per treatment) for a digestibility study (Exp. 2) were randomly allocated to 4 experimental treatments in a 2 × 2 factorial arrangement. Four dietary treatments were control ( CS ) based on corn and 44% CP soybean meal (ME: 2,850 kcal/kg CP: 18.5%) and CS-low energy/protein ( CSL ) (ME: 2,750 kcal/kg CP: 17.5%), with or without 0.05% β-mannanase enzyme. Hens were fed the experimental diets for 14 d for the digestibility study and 8 wk for the production study. Hen-day egg production ( HDEP ), weekly feed intake, FCR, and biweekly egg quality parameters were measured. Significant interaction on feed intake ( P < 0.01) was observed between energy/protein and enzyme. At 3, 6 and 8 wk, the feed intake and FCR of CSL with enzyme were significantly lower than those of CSL without enzymes. The main effects indicated that birds fed diets without inclusion of β-mannanase had higher feed intake than those fed diets with enzymes at 4, 7, and 8 wk. The inclusion of β-mannanase significantly increased ( P < 0.05) HDEP at 2, 3, 5, and 7 wk. However, there was no significant effect of nutrient density or enzyme supplementation on egg quality parameters. The digestibility study showed that the inclusion of β-mannanase significantly improved ( P < 0.01) apparent ileal digestibility of lysine, histidine and tryptophan in the diet. The results of these experiments indicate that supplementation of β-mannanase could reduce the feed intake and FCR and improve HDEP and apparent ileal digestibility of key amino acids in corn/soy diets fed to laying hens.

Dietary β-mannanase is reported to decrease antinutritional effects of β-mannans, such as the potential increase of body heat; however, its efficacy on broiler chickens raised under hot climatic conditions has not been investigated. Objective: To investigate the effects of dietary β-mannanase on growth performance, cloacal temperature, relative lymphoid organ weight, and blood characteristics of broiler chickens raised under hot climatic conditions. Methods: A total of 1,701 1-day-old Ross 308 broiler chickens were randomly allotted to one of three dietary treatments with nine replicates. A basal diet was prepared and added with β-mannanase at 0.05 or 0.10% inclusion levels. The experiment was conducted for 30 days. Average room temperature was 28.8 ± 1.74 ˚C and average relative humidity (RH) was 76.1 ± 11.49% during the experiment. Results: Growth performance of broiler chickens raised under hot climatic conditions was not affected by β-mannanase inclusion. Cloacal temperature decreased at the end of experiment (linear, p<0.05) with increasing inclusion levels of dietary β-mannanase. Increasing inclusion levels of β-mannanase tended to increase (linear, p=0.076) the relative weight of thymus, but had no effects on the relative weight of spleen and bursa of Fabricius. Blood characteristics were not influenced by dietary β-mannanase. Conclusion: Increasing inclusion levels of β-mannanase decrease cloacal temperature; however, it does not directly influence growth performance nor alleviates the heat stress of broiler chickens raised under hot climatic conditions. ResumenAntecedentes: Altas cantidades de carbohidratos no almidonosos en la dieta pueden aumentar la cantidad de materiales fermentativos en el intestino posterior, aumentando la producción de calor fermentativo.
