Effects of different amino acids on somatic embryogenesis of strawberry (Fragaria × ananassa Duch.)

Abstract: This study was conducted to optimize different types and concentrations of amino acids on somatic embryogenesis induction, development and maturation of leaf explants in strawberry cultivars ('Camarosa', 'Paros' and 'Kurdistan'). Calli derived from leaf sections were transferred onto MS medium with 1.0 mg/l 2,4-D ? 0.5 mg/l BAP supplemented with 0.0, 50, 100, 150 or 200 mg/l concentrations of proline, alanine and glutamine. Stimulation of embryogenesis and embryo development was strictly dependent on the type … Show more

“…1). Similarly, several reports confirmed that formation of SEs was enhanced by glutamine (Baskaran and Jayabalan 2009;Deo et al 2010;Gerdakaneh et al 2011). The role of picloram, TDZ and glutamine on SEs production has also been reported in other plant species (Tribulato et al 1997;Van Staden 2012, 2014).…”

“…Furthermore, combination of picloram, TDZ and glutamine induced high production of SEs (Table 1). The addition of an organic nitrogen form has a positive influence on somatic embryogenesis (Robichaud et al 2004;Zouine and Hadrami 2007;Gerdakaneh et al 2011). Glutamine has been shown to improve somatic embryogenesis in both monocotyledons and dicotyledons (Dhir et al 1991;Yin et al 1993;Ke et al 1996;Vani and Reddy 1996;Shrivastava and Chawla 2001).…”

“…Thus, it is usually recommended to take explants from plants grown under controlled conditions such as a growth room or greenhouse or from buds that flush from dormant shoots stored indoors. 7,37 To evaluate the best media for the shoot formation, the juvenile leaf explants of the three cultivars (Festival, Sweet Charlie, and Florida) were cultured on MS medium with TDZ at concentrations of 1, 2, and 3 mg/l, individually. Cultures were incubated at dark for a week, then transferred to light for at least three weeks.…”

“…Similarly, in strawberry when sucrose concentration was raised from 30 to 90 g/l callus fresh weight was decreased (Gerdakaneh et al 2010). According to Gerdakaneh et al (2009), low sucrose concentration is much more effective than high levels on callus fresh weight as the rise of sucrose concentration decreases callus fresh weight and this capacity could be attributed to an osmotic effect. In CAB-6P, the increase in sucrose concentration from 15 to 30 g/l led to lower callus induction frequencies.…”
