Given the differences in the dopamine neurotransmission between the shell and the core of the nucleus accumbens, as well as the differential involvement of these two domains in oral behaviour of rats, it was decided to determine whether or not dopamine Dj and/or dopamine D 2 receptors differentially direct oral behaviour in these two domains in rats. Intra-accumbens injections of the dopamine D, receptor agonist ( + )-6-chloro-7,S-dihydroxy-3-alIyl-1 -phenyl-2,3,4,5-letrahydro-l/7-3-benzazepine (SKF 82958: 5 jiig/0.2 /¿I), the dopamine D2 receptor agonist quinpirole (10 ju,g/0.2 ¡j. 1) and their combination were used to assess the role of these accumbens domains in jaw movements of rats. The present study shows that the combined administration of SKF 82958 and quinpirole into the shell, but not the core, of the nucleus accumbens produced a highly significant increase in jaw movements, when doses which per se were nearly ineffective, were injected. This effect was fully inhibited by prior administra tion of either the dopamine D [ receptor antagonist 7M + )-7-chloro-8-hydroxy-3-methyl-l-phenyl-2,3,4,5-tetrahydro-l //-3-benzazepine (SCH 23390: 0.5 ¿¿g/0.2 ¡M g) or the dopamine D 2 receptor antagonist (-)-sulpiride (25 ng/0.5 /xl) into the same region. It is concluded that dopamine D, and D 2 receptors in the shell, but not the core, of the nucleus accumbens are involved in jaw movements of the rat, providing the first piece of evidence that dopamine Dj and D 2 receptors in the shell of the nucleus accumbens mediate a particular behaviour.

Orofacial dyskinesia (OFD) is a disorder characterized by involuntary movements of the oral and facial muscles. OFD attacks can be elicited acutely in cats by local injections of dopaminergic agents into the anterodorsal part (r-CRM) of the caudate nucleus. Because the dopaminergic A8 cell group, being embedded in the retrorubral nucleus (RRN), gives rise to fibres which terminate in the r-CRM, two questions arose: (1) whether the A8 cell group forms part of the circuitry that directs and/or modulates OFD, and (2) whether GABA-ergic compounds in the RRN play a role in OFD, and if so, whether a pharmacological GABA-ergic intervention of the activity in the RRN modulates or mediates OFD. For this purpose, the activity of the RRN was manipulated with local injections of the GABA(A) agonist muscimol and antagonist bicuculline. These local injections into the RRN were subsequently combined with manipulations of dopamine transmission in the r-CRM with local injections of the selective DAi receptor agonist (3,4-dihydroxyphenylimino)-2-imidazoline. The present study shows that local injections of GABA-ergic compounds into the RRN do not elicit OFD attacks in cats, but can modulate oral behaviour elicited from the r-CRM. The latter effect is dose dependent and GABA-ergic specific.

Rats received haloperidol (1.0 mg/kg i.p.) or clozapine (10 mg/kg i.p.), twice daily for 4 weeks: vacuous chewing--recorded 26 h after the final injection--similarly increased in both groups. Three h later, the rats were challenged with dopaminomimetics, and automatically recorded jaw movements were analysed. Both apomorphine and a mixture of D1 and D2 receptor agonists (SKF 38393 resp. quinpirole) increased jaw movements in haloperidol-treated, but not clozapine-treated rats; SKF 38393 or quinpirole remained ineffective, when given alone. A fixed dose of quinpirole together with increasing doses of SKF 38393, but not a fixed dose of SKF 38393 together with increasing doses of quinpirole, produced a dose-dependent increase in jaw movements in otherwise non-treated rats, suggesting that the noted haloperidol-induced increase was due to a shift in the D1-D2 receptor balance towards a predominance of D1 receptors. This study presents a new animal model of tardive dyskinesia with predictive validity, good reliability and, especially, great efficiency.
