Effects of Enfonidipine Hydrochloride in Dogs with Experimental Supraventricular Tachyarrhythmia

Abstract: ABSTRACT. It is required not to increase the ventricular rate and to preserve the ventricular systolic function in treating supraventricular tachyarrhythmia (SVTA). The objective of this study is to investigate whether or not Efonidipine hydrochloride (EH), a T and L dual type Ca 2+ channel blocker, suppresses the increasing ventricular rate without reducing the ventricular systolic function using canine SVTA models by rapid atrial pacing (RAP) method. Clinically healthy fourteen beagles were used. The 14 dogs… Show more

“…The Ao-PEP and Ao-ET were considered susceptible to the effects of the HR, and an increased HR reduced the Ao-PEP and Ao-ET, whereas a decreased HR reduced the Ao-PEP and Ao-ET [ 20 , 25 ]. Therefore, the ratio of Ao-PEP to Ao-ET (Ao-PEP/ET) has been used as an effective index for the clinical evaluation of ventricular contractility, as with FS [ 7 , 8 ]. No significant change was observed in the Ao-PEP/ET in this study, suggesting that the decreases in the Ao-PEP and Ao-ET resulted from an increase in the HR.…”

“…Reduction of LAP is a desirable goal for drugs used to treat congestive heart failure in MVD. In general, calcium channel blockers are used for treatment of hypertrophic cardiomyopathy [5-7], atrial fibrillation [8-10], and systemic hypertension [11,12]. Amlodipine is a long acting 1,4-dihydropyridine calcium channel blocker, often used for treatment of systemic hypertension.…”

“…Under general anesthesia, a pacing lead was implanted in the animals in accordance with the guidelines provided by Fukushima [9] and Gaspo et al [12]. In the present study, for stimulation of the atrium by RAP (2 mV, 390 bpm), artificial tachycardia was immediately triggered and maintained in the dog, and hemodynamics resembling the spontaneously developed similar to AF were obtained [9, 12] (Fig.…”
