Effects of Eosinophilotropic Cytokines on Differentiation of an Eosinophil Cell Line, YY-1

Abstract: Differentiation of an eosinophilic cell line, YY-1, was induced by treatment with 0.3 mM butyric acid (BA). The effects of eosinophilotropic cytokines, such as IL-3, IL·5, and GM-CSF on BA-induced differentiation, especially in the chemotactic response to ECF, was examined. Five STO-2-derived ECFs, IL·5, C5a, and fMLP were used as eosinophil chemoattractants. The effects of the cytokines on the chemotactic response of YY-1 greatly differed according to the ECF used. It was thus suggested that heterogeneity in … Show more

“…However, its expression is downregulated on mature human eosinophils when treated the same cocktail of cytokines (Gregory et al, 2003). Although the receptors are found on a wider range of cells, administration of IL-3 and GM-CSF has been shown to promote eosinophil production in animal models, as well as in clinical trials (Sanderson, 1992; Ueno et al, 1994; Takamoto and Sugane, 1995). …”

“…The differential responsiveness of eosinophils isolated from patients or normal controls is still a puzzle. n-BA -induced maturation of YY-1 cells [21,22] leads to an increased IL-18R expression and the treated YY-1 cells could respond to IL-18. A fraction of peripheral eosinophils from HES patients could also be activated [28].…”

“…In addition, it is difficult to obtain sufficient eosinophils for functional analysis due to their small numbers in the peripheral blood cells. We therefore checked expression of IL-18R in the human eosinophilic cell lines, YY-1 [21,22] and EoL-1 [23] and other human cell lines including B cells (NALM6, RAMOS, DND39), myeloma cells (U266), T cells (Jurkat), monocytic cells (U937) and basophilic cells (KU812). As shown in Fig…”
