Effects of Ghrelin on the Apoptosis of Rheumatoid Arthritis Fibroblast-Like Synoviocyte MH7A Cells

Abstract: Ghrelin is a circulating peptide hormone, which involved in promoting feeding and regulating energy metabolism in human and rodents. Abnormal synovial hyperplasia is the most important pathologic hallmark of rheumatoid arthritis (RA), which is characterised by tumor-like expansion. Existing studies indicated that there may exist some relation between the decreased ghrelin and the abnormally proliferating synovial cells in RA. Therefore, the aim of this study is to explore the apoptotic effects of ghrelin on MH… Show more

“…No obvious association was found between ghrelin and the SLEDAI scores in obese and non‐obese SLE patients. Ghrelin levels were significant lower in obese SLE patients with arthritis than those without, which was in accorded with some previously researches that ghrelin was decreased in rheumatoid arthritis and osteoarthritis 29–31 . A previous study found that ghrelin can reduce the expression of TNF‐α 30 .…”

[29][30][31] A previous study found that ghrelin can reduce the expression of TNF-α. 30 Besides, ghrelin can decrease IL-1β induced MMP-3, MMP-13, ADAMTS-4 and ADAMTS-5 increasing in chondrocytes.…”

“…The human synovial cell line MH7A was purchased from Riken Cell Bank (Ibaraki, Japan) and cultured in RPMI-1640 medium (Hyclone, USA) with 10% fetal bovine serum (FBS, Thermo, USA) and 1% penicillin/streptomycin (P/S, Hyclone, USA) at 37 °C in 5% CO 2 humidified atmosphere [ 23 ]. Primary synovial cells from a rheumatoid arthritis patient were kindly provided by Dr. Qingwen Wang from Peking University Shenzhen Hospital, Shenzhen, China.…”

“…Recent research suggests that as the main synovial cell type in synovial tissue, rheumatoid arthritis synovial fibroblast (RASF) is involved in maintaining the homeostasis of the synovial environment 7. Studies have reported that the transformation of biological characteristics of RASFs in the progression of RA is an early pathological change 8 9. Research based on RASFs has become a direction for RA treatment.…”
