Daphnia spinulata Birabén, 1917 is an endemic cladoceran species, frequent in the zooplankton communities of the shallow lakes of the Pampean region of Argentina. These lakes have varying salinity levels and, being located in agricultural areas, are frequently subject to pesticide pollution. This study aimed to determine the effects of the herbicide glyphosate (Panzer Gold®) in combination with different salinity levels on the biological parameters of D. spinulata and its recovery ability after a short exposure. Three types of assays were performed: an acute toxicity test, a chronic assessment to determine survival, growth and reproduction, and recovery assays under optimal salinity conditions (1 g L − 1 ). The LC 50 -48h of glyphosate was 7.5 mg L − 1 (3. 15-11.72). Longevity and the number of offspring and clutches were signi cantly reduced by glyphosate and high salinity exposure, whether in isolation or combined. The timing of the rst offspring did not recover after glyphosate exposure. Our results reveal that D. spinulata is sensitive to the herbicide, which causes stronger negative effects in conditions of higher salinity. Further research is needed to shed further light on the sensitivity of this cladoceran to glyphosate and its variability under other interactive stress factors.

Many herbicide products are commonly used in agricultural areas to prevent and eliminate weeds. Contamination from these toxicants in water might affect aquatic organisms not only in the active stage, but also in the diapause stage. To test the effect of herbicide on the resting eggs of zooplankton, we prepared two rice fields: one field without the application of pesticides (RF−NPA) and one with the application of pesticides (RF−PA) in a sampling year. We conducted a hatching experiment for 30 days. Twenty–four taxa of zooplankton were found. Sixteen species of these were rotifers, seven species were cladocerans and one taxon was an unidentified nauplius copepod. The species richness of zooplankton between RF–NPA (17 taxa) and RF–PA (16 taxa) was close, but species compositions between RF–NPA and RF–PA were different, indicated by the similarity index of 0.545. Lecanidae was the most diverse family of rotifers in both rice fields with nine species, while Chydoridae was the most diverse family of cladocerans (four species). The total abundance of zooplankton of RF−NPA was higher than RF−PA with 1,897 and 1,286 individuals, respectively. The Shannon–Wiener diversity index (H´) and Pielou’s evenness (J) in RF−NPA were higher than in RF−PA. The high species richness of zooplankton in both rice fields occurred on days 18 to 30. On the other hand, the highest abundance was recorded on day 18 for RF−NPA and on day 24 for RF−PA. The non-metric multidimensional scaling (NMDS) demonstrated significant differences in zooplankton community composition between RF–NPA and RF–PA (p < 0.05; ANOSIM test). According to the diversity indices, the RF–NPA has more diversity than the RF–PA, which might be a result of herbicide application in the sampling year. This study suggests that the toxicity of glyphosate should be a concern in terms of the biodiversity of rice field ecosystems.

Roundup ® is the most used glyphosate-based herbicide. During agricultural use it may directly contaminate existing aquatic ecosystems, posing severe concerns for the safety of nontarget terrestrial and aquatic organisms. We investigated the outcome of exposure to different concentrations of glyphosate in Roundup on cyst hatchability, toxicity, and teratogenic effects in the aquatic crustacean Artemia salina that inhabits diverse types of salt waters and, as a filter feeder, carries a greater risk of being exposed to pollutants. We found that exposure to 144 and 288 μg/ml glyphosate in Roundup resulted in cysts unable to complete diapause, and hatchability was completely inhibited during all exposure times tested (17-48 h). A glyphosate concentration of 288 μg/ml in Roundup was lethal to A. salina nauplii, and the lower concentrations (9, 18, 36, 72 μg/ml) had no significant effects on viability. In addition, sublethal and environmentally safe concentrations of glyphosate (0.72 μg/ml) in Roundup affected the early development of A. salina nauplii, with significantly decreased body lengths and reduced widths of the tail, abdomen, and head. The increased level of catalase activity observed in nauplii exposed to 0.72 μg/ml glyphosate for 24 h and those exposed to 7.2 and 72 μg/ml glyphosate for 48 h may be linked to excessive reactive oxygen species levels that had been induced by Roundup. In conclusion, Roundup containing >72 μg/ml glyphosate totally inhibited hatching of cysts and exerted toxic effects on A. salina nauplii. The increased prevalence of developmental defects in the nauplii observed at 0.72 μg/ml glyphosate signifies possible teratogenicity of Roundup exposure even at environmentally relevant concentrations of glyphosate, possibly due to disturbance of the antioxidant defenses, which needs further investigation.
