Effects of GnRH analogues, `add-back' steroid therapy, antiestrogen and antiprogestins on leiomyoma and myometrial smooth muscle cell growth and transforming growth factor-beta expression

Abstract: The objective of this study was to elucidate the biological significance of GnRH and antiprogestins and antiestrogen in leiomyoma and their interactions with ovarian steroid 'add-back' therapy. Leiomyoma and myometrial smooth muscle cells (LSMC and MSMC) were isolated and exposed to GnRH agonist (leuprolide acetate, LA), 17beta-estradiol (E2), medroxyprogesterone acetate (MPA), GnRH antagonist (Antide), estrogen antagonist, ICI182780 (Fulvestrant) and progesterone antagonists RU486 (Mifepristone) and ZK98299 (… Show more

“…Tumor growth is strongly dependent on estrogen and progesterone, and both have a mitogenic effect on leiomyoma cells (5). Estrogen and progesterone seem to have a mitogenic effect on leiomyoma cells and to act by influencing (directly and indirectly) other hormones and a large number of growth factors, cytokines and apoptotic factors (6).…”

“…When given for 3-6 months, mifepristone reduced UL size and symptoms and induced amenorrhoea in 63-100%of women [22] . Mifepristone induces a significant decrease in uterine volume, a significant reduction in Pg receptors content without affecting the estrogen receptor level [8,23] . Under in vivo conditions RU486 also altered the rate of DNA synthesis and growth factors and proteases expression in MSMC or LSMC primary cultures [8] .…”
