Electrophoretic techniques, used to separate mixtures of electrically charged particles, are widely used in science. One of these techniques, native protein electrophoresis in an agarose gel, is applied in human and veterinary medicine. Changes in the proportions of individual protein fractions correspond to significant changes in the physiology of the body. Although the pattern obtained by electrophoretic separation rarely indicates a specific disease, it provides valuable information for the differential diagnosis. Decades of research on the types of patterns obtained in the case of particular diseases have led to the accumulation of substantial knowledge. The paper presents the available information on this topic. Serum protein electrophoresis is recommended in cases of increased levels of total protein in order to reveal the nature of the process. The basic information which can be obtained from electrophoretic separation includes the immune status of the organism. Both increased antigenic stimulation and immunodeficiency are clearly visible in electropherograms. Moreover, the level of heterogeneity of the corresponding protein fractions can help to distinguish between infectious diseases and cancer -multiple myeloma -the latter producing a homogeneous immunoglobulin fraction. Analysis of other protein fractions helps to detect or confirm an ongoing inflammatory process and provides information regarding liver function. Even when the concentration of total protein is within the reference range, this analysis can be recommended as a basic laboratory test.

An acute phase index based on a combination of acute phase proteins which permitted monitoring the response to therapy of canine leishmaniasis was developed and evaluated in this study. Six dogs naturally infected by Leishmania infantum were treated with meglumine antimoniate (Glucantime  , Merial, Lyon, France) 100 mg/kg/day sc, given concurrently for 20 days with allopurinol (Zyloric  , Glaxo Wellcome, Madrid, Spain) 30 mg/kg/day po and then, allopurinol alone for one month at the same dosage. Blood samples for acute phase proteins were obtained on different days before and after the beginning of treatment and two groups of indexes were calculated: (1) Indexes that combined one positive and one negative acute phase protein and (2) Indexes that combined two positive and one negative acute phase proteins. All calculated indexes were significantly higher in animals with leishmaniasis compared with clinically healthy dogs (n = 8) and a decrease was observed in all dogs tested during the treatment. Indexes that combined C-reactive protein (CRP) and ceruloplasmin (CP) with other proteins showed greater percentages of decrease that were statistically significant. Among these, the index CRP*CP/Alb was selected as the optimum since it showed a larger and faster decrease compared with the others as well as with individual proteins alone. These results would support the use of selected acute phase indexes, especially the CRP*CP/Alb index, to suspect about a leishmaniotic dogs and to monitor their response to treatment.

Comparison of serum protein electrophoretic pattern in cows and small ruminants

Determination of the physiological electrophoretic patterns in animals is very useful for clinicians in diagnosing healthy and sick animals. The objective of this study was to investigate the serum protein electrophoretic pattern in cows, sheep, and goats in order to evaluate the differences in the size and number of protein fractions between the evaluated ruminant species. Ten adult multiparous high-yielding dairy cows, 10 adult female sheep and 10 adult female goats were included in this study. All the evaluated animals were clinically healthy. Serum was analyzed for total serum protein concentrations, and for the relative and absolute values of protein fractions with calculation of albumin/globulin ratios. Serum protein fractions were separated by zone electrophoresis on buffered agarose gel. Serum protein electrophoresis identified 6 distinct bands, comprising albumin, alpha 1 -(α 1 ), alpha 2 -(α 2 ), beta 1 -(β 1 ), beta 2 -(β 2 ), and gamma-(γ) globulins in cows. In sheep, serum proteins exhibited 6 fractions: albumin, α 1 -, α 2 -, β-, γ 1 -and γ 2 -globulins. In goats, serum proteins were separated into 5 fractions: albumin, α 1 -, α 2 -, β-and γ-globulins. Significant differences in the relative as well as absolute means were found for the albumin/globulin ratio and most of the protein fractions, except γ-globulins. No significant differences were found in the concentration of total proteins. These results describe the marked species differences in most of serum protein fractions between the evaluated groups of animals, and contribute to the current knowledge about the physiological electrophoretic pattern of serum proteins in ruminants, which can be used for diagnostic purposes.
