Effects of hydroethanolic extracts of Terminalia chebula and Thymbra spicata on ram fresh semen under normal and oxidative stress conditions

Abstract: The objective of this study was to evaluate protective effects of hydroethanolic extracts of Terminalia chebula and Thymbra spicata on viability, lipid peroxidation (LPO) and DNA integrity of ram fresh semen under normal and oxidative stress (OS) conditions. Antioxidant activities of different concentrations of Terminalia chebula and Thymbra spicata extracts were evaluated with DPPH assay. Semen samples were taken from three fertile adult rams. After diluting semen with Tris‐base extender, different concentrat… Show more

“…Mitochondrial enzymes are capable of reducing tetrazolium to insoluble formazan (Kime et al., 2001). In experiment 1, to evaluate sperm viability, MTT assay was performed according to a method described by Mohammadi and Soltani (2021). The MTT solution was prepared as a 5 mg/ml stock solution in Dulbecco's phosphate‐buffered saline (DPBS), filtered and kept for no more than two weeks in the dark at 4°C.…”

“…As a diluent, a Tris base extender was used in this study (Mohammadi & Soltani, 2021). 0.5 g fructose was mixed with 3.63 g tris‐based, 1.99 g citric acid, 6% glycerol and 20% egg yolk in 100 ml distilled water as the extender.…”

“…Tetrazolium can be reduced by mitochondrial enzymes to an insoluble formazan 29 . Using the protocol outlined by Mohammadi and Soltani 30 , the MTT test was performed to evaluate the vitality of diluted fresh sperm. Dulbecco's phosphate-buffered saline (DPBS) was used to generate a 5 mg/mL MTT solution, which was then filtered and stored at 4 °C in the dark for a maximum of 2 weeks.…”
