Effects of Ketanserin – a Serotonin Receptor Antagonist – on Placental Blood Flow, Placental Weight and Fetal Weight of Spontaneously Hypertensive Rats and Normal Wistar Kyoto Rats

Abstract: Recently, ketanserin (serotonin receptor antagonist) has been reported to be beneficial in the treatment of preeclampsia. In this study, we investigated the effects of ketanserin on the placental blood flow, fetal weight and placental weight in spontaneously hypertensive rats (SHR) and normotensive Wistar Kyoto rats (WKY). We measured the rats’ placental blood flow by monitoring the clearance of hydrogen gas generated by electrolysis. In ketanserin (12, 24, 48 mg/kg)-treated SHR, systolic blood pressure was si… Show more

“…Reduced fetal weight has been noted previously in the SHR (Ahokas et al 1986) but placental weights were not reported. However, two Japanese studies reported no difference in either fetal or placental weights between SHRs and WKY rats (Furuhashi et al 1991(Furuhashi et al , 1992. Unfortunately, comparisons of results from different centres using SHRs and WKY rats can present difficulties because it is now known that not all SHRs or WKY rats constitute single inbred strains.…”

“…It is well known that antihypertensive treatment for pre eclampsia results in a concomitant significant decrease of uteroplacental blood flow, which in turn adversely affects the oxygen supply to the fetus. In this study, we measured the placental blood flow in the PGK-treated rats with the clearance method using electroiytically generated hydro gen in order to investigate the effects of PGH on the pla cental blood flow as previously reported [21,22], The effect of exogenous PGH reduced placental blood flow in the SHR groups; on the other hand, the placental blood flow increased in the WKY 0.5-mg-treated group ( fig. 3).…”

“…We used a model of the instrument for evaluating the clearance of hydrogen gas generated by electrolysis made by Biomedical Science Co. (Kanazawa, Japan) as reported in our previous study [ 17,18].…”
