Effects of local administration of tiludronic acid on experimental periodontitis in diabetic rats

Abstract: Local administration of TIL promoted a protective effect against tissue destruction in EP in diabetic rats, and the dosage of 3 mg/kg of TIL promoted the best results regarding its antiresorptive and anti-inflammatory effects.

“…The X-ray generator was operated at 50 kV with a source current of 500 µA and a 0.5 mm aluminum filter. A volume of interest from the apexes of all roots of the mandibular first molar (M1) up to the roof of the furcation of M1, touching the roots surfaces (CT-Analyzer; Bruker, Belgium) was selected for bone volume measurement, as previously described 37,38 . The bone volume/tissue volume (BV/TV), porosity, trabecular number (Tb.N) and trabecular separation (Tb.Sp) were analyzed in the volume of interested area with CtAn (Bruker, Belgium).…”

“…Administration of cannabinoid‐2 receptor agonist Hu‐308 into the gingival tissue or interdental space of rats prevented bone resorption and decreased the gingival production of inflammatory mediators in rats with LPS‐induced periodontitis . Application of tiludronic acid into the gingival tissue promoted a protective effect against tissue destruction in EP of diabetic rats . Our study similarly followed the protocol as in these studies, and presented the advantage of avoiding possible systemic adverse effects of rhPGRN.…”

“…Local application of anti-inflammatory agents has been employed in some periodontitis model studies. [39][40][41] Administration of cannabinoid-2 receptor agonist Hu-308 into the gingival tissue or interdental space of rats prevented bone resorption and decreased the gingival production of inflammatory mediators in rats with LPS-induced periodontitis. 40 Application of tiludronic acid into the gingival tissue promoted a protective effect against tissue destruction in EP of diabetic rats.…”

The rat models, used in the reviewed publications were: ligature-induced [5,13,[15][16][17][18][19], streptozotocin-induced [2,3,20,21] and immune complexes-induced [22] experimental periodontitis; 3-wall periodontal defect in Wistar rats [9,14],osteoporotic rats periodontal fenestration defect model [4], apical periodontitis, induced in Wistar rats by pulp exposure to oral cavity [23], altered functional loading model with use of a hard and soft diet and the posterior bite-blocks in rats [24], as well as rats stress exposures in different modes [25], experimental rat model of surgically created chronic acid reflux esophagitis [26].
