Effects of lupus‐inducing drugs on the B to Z transition of synthetic DNA

Abstract: Five drugs associated with systemic lupus erythematosus were studied for their effect on the salt-induced right-handed (B) to left-handed (Z) transition of poly (dG-me5dC) . poly(dG-me5dC). Using circular dichroism spectroscopy, procainamide and hydralazine were found to reduce the midpoint of B to Z transition from 0.8M NaCl to 0.5M NaCl and to increase the rate of this transition at 1M NaCI. Isoniazid and D-penicillamine had less effect on the midpoint of transition and practically no effect on the kinetics… Show more

“…In contrast to the poor immunogenicity of B-DNA, Z-DNA is immunogenic. PA can facilitate the conversion of B-DNA to Z-DNA (13) and, thus, may disrupt nuclear structure, with the potential for exposure of histones in an immunogenic form. In this study, 34 patients (65.4%) had IgM and/or IgG antibodies to total histones.…”

“…5-azaC is incorporated into newly synthesized DNA, where it covalently binds the methyltransferase that methylates cytidine in the newly synthesized DNA strand when 5-methyldeoxycytidyldeoxyguanosine is recognized on the parent strand (18). Pca and Hyd bind DNA (19)(20)(21), and may interfere with the action of the methyltransferase, resulting in hypomethylation of the newly synthesized strand.…”

“…Mercuric chloride can induce antifibrillarin antibodies in susceptible mouse strains (79), and mercury ions will bind fibrillarin, potentially altering its antigenicity (80). Hydralazine, procainamide, and to a lesser degree, isoniazid and D-penicillamine change the usual right-handed helical configuration of DNA (B DNA) to a more immunogenic left-handed helical configuration (Z DNA) in vitro (81). This conversion has been proposed as a mechanism contributing to the development of anti-DNA antibodies in druginduced lupus (81).…”
