Effects of Methotrexate on Vascular Endothelial Growth Factor, Angiopoietin 1, and Angiopoietin 2 in Nasal Polyps

Abstract: MTX may inhibit the growth of NPs via local regulation of VEGF, Ang-1, and Ang-2 protein levels. We suggest that MTX can be used to treat NPs.

“…With the increased VEGF expression, vascular permeability is increasing, the invasion of inflammatory cells, fibroblasts and endothelium cells would induce the formation of granulation tissue. Meanwhile, because of increased plasma extravasation, extracellular matrix accumulation and tissue edema, local increased force induces epithelial rupture, slowly forming nasal polyps [14,15]. In the current study, the positive expression of VEGF in nasal polyps was 75%, while low expression in inferior turbinate tissue, indicating the important role of VEGF in the development of nasal polyps.…”

“…11 Another aspect is the overexpression of vascular endothelial growth factor (VEGF) in nasal polyps and its relevance for polyp formation that has gained increasing attention. 12 Previously, we and others reported about increased VEGF levels in nasal polyps 13,14 potentially associated with tissue edema, proliferation, and apoptosis. 15 Moreover, survivin up-regulation by VEGF is well known from several types of tissues.…”

“…Moreover Methotrexate inhibits in vitro corneal vascular endothelial cell proliferation and blocks VEGF-and bFGF-induced corneal neovascularization in vivo (Hirata et al, 1989). Others also demonstrated significant decrease in serum levels of VEGF after 2 and 6 months of treatment with low dose methotrexate and cyclophosphamide in patients with metastatic breast cancer (Colleoni et al, 2002) .In addition, neoadjuvant multidrug chemotherapy including high-dose methotrexate modifies VEGF expression in osteosarcoma (Rossi et al, 2010;Park et al, 2011).…”
