Effects of nomifensine in vitro on uptake of 5‐hydroxytryptamine and dopamine into human platelets.

Abstract: 1The effect of nomifensine on the uptake of 5-hydroxytryptamine (5-HT) and dopamine (DA) into human platelet-rich plasma has been studied. 2 A significant inhibition of DA uptake was observed at nomifensine 10-6M. 3 5-HT uptake was significantly inhibited only at nomifensine 10-4M or more. 4 These results are consistent with those from previous animal experiments and suggest that the human platelet may be used as a model for studying DA uptake as well as that of 5-HT.

“…Nomifensine, an inhibitor of monoamine reuptake preferring the dopamine transporter (Ehsanullah and Turner 1977), while not a drug of abuse per se, has been found to be sufficiently reinforcing to establish lever-pressing behavior in rats (Carlezon et al 1995). Nomifensine competes with, and can displace cocaine for its binding site on the dopamine transporter (Missale et al 1985).…”

“…A further 3 patients (2 males, one female) with established HC were matched with normal subjects, and platelet DA and 5-HT uptake was studied. Platelet monoamine uptake was measured by the method described elsewhere (Ehsanullah and Turner, 1977).…”

“…I would like to supplement the studies of Ehsanullah & Turner (1977) with some studies of my own regarding the effect of nomifensine on 5-hydroxytryptamine (5-HT) uptake in human blood platelets. Time does not allow me to describe the method in detail (see Lingjaerde, 1976).…”
