Effects of obesity and diabesity on heart rhythm in the Zucker rat

Abstract: Obesity and type 2 diabetes mellitus are risk factors for hypertension, coronary heart disease, cardiac arrhythmias including atrial fibrillation, heart failure and sudden cardiac death. The effects of obesity and diabesity on heart rhythm were investigated in the Zucker diabetic fatty (ZDF) and Zucker fatty (ZF) compared to the Zucker lean (ZL) control rat. In vivo biotelemetry techniques were used to assess the electrocardiogram and other cardiac and metabolic parameters. ZDF rats were characterized by age‐d… Show more

“…Finally, gap junction protein Connexin Cx45 is important for electrical signal conduction between cardiac myocytes and heart rhythm [ 51 ]. In this experiment, the expression of Cx45 was reduced in ZF and modestly reduced in ZDF compared to ZL ventricle tissue, which may have implications for disturbances in heart rhythm, which have been reported in a previous study [ 52 ]. Interestingly, it has been reported that mRNA expression levels for Gja7 (Cx45) was significantly increased in SAN from diabetic heart compared to controls [ 53 ].…”

“…In conclusion, increased neuronal activation in sympathoexcitatory brain regions has been demonstrated for the first time in an animal others. 67,68 Current therapies that target sympathoexcitation in type 2 DM have reduced effectiveness compared to non-DM individuals. 4 Our findings suggest that pharmacotherapies 5 targeting the central source of cardiac sympathoexcitation in type 2 DM might be an alternative treatment strategy.…”

“…Recent in vivo biotelemetry experiments have demonstrated that HR was significantly reduced in ZDF compared to ZF and ZL control rats in the absence of changes in body temperature and physical activity [47]. Age related changes in PQ intervals, QRS intervals, QT and QTc intervals between different groups were also reported.…”
