Effects of pentoxifylline and polymyxin B on the acute‐phase‐response to Escherichia coli endotoxin in dwarf goats

Abstract: The purpose of this study was to assess whether polymyxin B together with pentoxifylline, had beneficial effects on the acute-phase-response to E. coli endotoxin in the dwarf goat (n = 6). Polymyxin B partly neutralizes E. coli endotoxin by forming inactive polymyxin B-lipopolysaccharide (LPS) complexes; pentoxifylline has been reported to suppress the LPS-induced production of tumour necrosis factor (TNF-alpha). E. coli LPS (0.0067 microgram/kg/min over 30 min) induced fever, tachycardia, inhibition of rumen … Show more

“…In the present study, we demonstrate that LPS from intestinal Gram‐negative bacteria may play a role in the activation of joint inflammation in which immune responses to pathological antigens are involved because oral administration of the endotoxin from E. coli , after AIA had been subsided, reactivated the joint inflammation. The reactivation of AIA was blocked by PMB that neutralized the endotoxin [15]. The role of LPS given p.o.…”

“…To investigate whether PMB that neutralizes LPS [15] can suppress the reactivation of AIA by the endotoxin, we gave the antibiotic orally immediately before administration of LPS. As shown in Fig.…”

“…The LPS active site lipid A was also effective in stimulating the existing joint inflammation. The reactivation of CIA by LPS and lipid A was blocked by PMB that neutralized these Gram negative bacterial cell wall components (van Miert et al ., 1987), supporting the role of LPS and lipid A in the recurrence of joint inflammation. The reactivation of CIA by LPS was also confirmed by histologic changes in joints of LPS‐treated mice showing marked oedema of synovium which had proliferated by the day of the endotoxin challenge, new formation of fibrin, and infiltration of many neutrophils accompanied with mononuclear cells, while control untreated animals failed to have such histologically acute inflammatory features.…”

“…Treatment with polymyxin B to attenuate clinical endotoxaemia is frequently used in adult horses for conditions such as strangulating intestinal lesions, pleuropneumonia, and postpartum metritis, with beneficial effects (34). The effects of intravenous applications of pentoxifylline and polymyxin B on the acute-phase-response to Escherichia coli endotoxin in dwarf goats have been evaluated (29). However, no study has been carried out on the clinical impact and anti-inflammatory effects of polymyxin B alone on endotoxaemia in sheep.…”
