Effects of Propylthiouracil Treatment on Antioxidant Activities in Blood of Toxic Multinodular Goiter Patients

Abstract: Erythrocyte, serum and plasma antioxidant activities and the effects of propylthiouracil (PTU) treatment on these activities were studied in patients with toxic multinodular goiter. The activities of the erythrocyte antioxidant enzymes (glucose-6-phosphate dehydrogenase, catalase, Cu/Zn-superoxide dismutase, selenium (Se)-dependent glutathione peroxidase and glutathione reductase) and the levels of erythrocyte Se, serum ceruloplasmin and plasma malondialdehyde were significantly higher while serum vitamin E, p… Show more

“…2 The effect of PTU on lipid peroxidation has been investigated previously. PTU reduced the extent of lipid peroxidation in both in vivo 24,25 and in vitro 6 studies with this agent. Moreover, PTU, as do other thionamides, has an immunosuppressive action that is beneficial for the treatment of Graves' disease.…”

“…Hyperthyroidism enhances ROS generation and produces changes in antioxidant systems of various tissues [34]. There are also conflicting reports on the immunosuppressive and free radical scavenging properties of anti-thyroid drugs [35][36][37]. To eliminate the influence of abnormal levels of thyroid hormones, the Graves' patients were included in this study after the achievement of stable euthyroidism by use of the same anti-thyroid drug and treatment protocol.…”

“…Firstly, it should be noted, that prooxidant activity of Se was shown to cause upregulation of Se-independent antioxidant enzymes, such as catalase and superoxide dismutase, in different biological systems, and these observations come from plants [169,170], yeast [171], fish [172][173][174][175][176] and, importantly, human cancer cell lines [50,177]. Secondly, such adaptive stress response, specifically with respect to Se-containing enzymes, is also well known to occur following acute and chronic injury 29 human studies [178][179][180][181][182][183][184][185][186][187][188][189][190][191][192][193][194][195][196] showing that selenoproteins (including GPx and Sepp1) are upregulated in the response to oxidative stress from their baseline levels without any change in Se supply and availability, i.e. as a consequence of adaptive response [168].…”
