Prostaglandin F2α (PGF2α) incorporated in Silastic‐PVP tubes 2 cm long and one end sealed induced temporary sterility when placed intrascrotally in fertile male rabbits. A significant reduction in sperm number per ejaculate, testicular weight and the number of epididymal sperm accompanied the incidence of sterility which occurred 2–5 weeks after starting the treatment and persisted for 6–10 weeks. During the experimental period, no significant deviation in libido or ejaculate volume compared to sham controls was observed. Prostaglandin F2α might have a local effect on the testis and epididymis, causing disintegration and depletion of the sperm reserve resulting in temporary sterility.

In adult male rats, bilateral intratesticular administration of 20 μg PGA1 or 20 μg indomethacin significantly reduced the acute stimulation of plasma testosterone levels by concomitantly injected LH, while treatment with the same dose of PGF2α was without effect. After six days of treatment with indomethacin, plasma testosterone levels were reduced to approximately 1/3 of the control levels. Intratesticular administration of 10 μg LH per testis per day produced, after six days, a precipitous decline in plasma testosterone levels and a decrease in the weight of the testes. This was probably due to desensitization of Leydig cells by these high doses of gonadotropin. Administration of PGF2α together with LH caused a further decrease in testicular weight and a significant reduction in the weights of the prostate and the seminal vesicles.

ONO-802, a synthetic E1 prostaglandin, was administered intravaginally via pessaries to Dutch belted rabbits at doses of 250, 62.5, and 12.5 micrograms/kg on days 6 through 18 of gestation. Rabbits in a vehicle control group were treated with pessaries that did not contain ONO-802 during the same period. Another group of animals remained untreated throughout gestation. Necropsies were performed on rabbits found dead and on those killed on gestation day 30. Body weight, food and water consumption, and clinical signs were monitored during the experiment. Major organs were weighed when the dams were necropsied on gestation day 30, and litter and fetal data were collected. Abortion and maternal deaths occurred in drug-treated groups. Body weight gains and food and water consumption were adversely affected by treatment particularly at the 250 and 12.5 micrograms/kg dose levels. Wastage (postimplantation loss) was significantly increased among treated groups (all dose levels), while other litter and fetal parameters were unaffected. ONO-802 was not teratogenic at maternal and embryotoxic dose levels.
