Effects of pulse methylprednisolone on inflammatory mediators in peripheral blood, synovial fluid, and synovial membrane in rheumatoid arthritis

Abstract: Objective. To establish whether the clinical efficacy of pulse methylprednisolone (MP; 1,000 mg intravenously) is related to the modulation of proinflammatory cytokines within the peripheral blood, synovial membrane, or synovial fluid compartments.Methods. Eighteen patients with active rheumatoid arthritis (RA) were studied. Peripheral blood (11 patients) and knee synovial fluid (9 patients, 10 knees) were obtained before and at 4 and 24 hours after MP therapy. Interleukin-lp (IL-lp), IL-8, and tumor necrosis … Show more

“…However, the current study is limited in its capacity to evaluate the complete implication of DCs in the pathogenesis of RA: the small number of patients, variability in patient's factors at study entry (disease duration, previous DMARD treatment [46,47]). It has to be continued in order to allow statistical correlations and to prove the importance of adhesion and co-stimulatory molecules expression on DCs as targets for future immunotherapies.…”

“…Annealing temperatures were 55°C for Fc␥RI and GAPDH, 60°C for Fc␥RIIa and Fc␥RIIb, and 62°C for Fc␥RIII. The PCR reaction was stopped after 20,23,26,29,32,35,38,41, and 44 cycles at the very end of the extension phase (72°C), and 5-l samples were obtained.…”

“…Sixty-one outpatients presented for management of RA to a specialist clinic. Of these, 20 had a flare of their disease, as previously defined (40), upon study entry. The Health Assessment Questionnaire (HAQ) (41) was administered to these 20 patients and to the HSCT group before treatment.…”
