Effects of RuPeng15 Powder (RPP15) on Monosodium Urate Crystal-Induced Gouty Arthritis in Rats

Abstract: RuPeng15 Powder (RPP15) is a herbal multicompound remedy that originates from traditional Tibetan medicine and possesses antigout, anti-inflammatory, and antihyperuricemic properties based on the traditional conceptions. The present study was undertaken to evaluate the therapeutic effect of PRP15 in rat gouty arthritis induced by monosodium urate (MSU) crystals. In the present study, we found that treatment with RPP15 (0.4, 0.8, and 1.2 g/kg) in rats with gouty arthritis induced by MSU crystals significantly a… Show more

“…Usually, NF-κB binds to the inhibiting protein, IκB and is localized in the cytoplasm. Certain stimuli, including MSU crystals, can cause IκB degradation and NF-κB translocation into the nucleus where it regulates the transcription of various target genes [18], [19], [20], [21], [22], [23], [24]. In this study, serum levels of IL-1β and TNF-α increased significantly in response to MSU crystals.…”

“…Recently, Rohde et al (2018) have provided strong evidence for the existence of a fully functional TNF-α/TNF-α receptor system in birds with an ability to induce NF-κB expression. Although the role of this cytokine in MSU-induced arthritis models has been confirmed in mammals (Liu-Bryan et al, 2005;Kou et al, 2015;Dhanasekar and Rasool, 2016), to the best of our knowledge this is the first time that its involvement in an avian model of this type of arthritis has been demonstrated. As previously stated, among different drug combinations, the administration of meloxicam (intramuscularly) + hydrocortisone (topically) showed better results (especially on TNF-α levels) followed by Groups T4 and T6.…”

“…Since 1962 when Faires and McCarty provoked inflammatory reactions in the knee joint of dogs and men injected with monosodium urate (MSU) microcrystals, the inflammatory nature of this model has been frequently shown in other animals, especially rodents. For instance, inflammatory cell infiltration and elevated expressions of nuclear transcription factor-κB p65 (NF-κB p65) in syn-Acta Veterinaria Hungarica 67, 2019 ovial tissues accompanied by increased levels of tumour necrosis factor-α (TNFα) and different interleukins (ILs) in the synovial fluid or serum of rats and mice have been reported (Liu-Bryan et al, 2005;Kou et al, 2015;Dhanasekar and Rasool, 2016). Although MSU-induced arthritis in birds is also an inflammatory process, knowledge on the possible changes in inflammatory cytokines is scarce in avian species.…”

“…In contrast, overexpression of TNF-α and IL-1β can directly stimulate the NF-κB pathway, lead to a positive feedback loop, then amplify the inflammatory response and cause joint damage [30][31][32][33][34]. Usually, NF-κB attaches to the inhibiting protein, IκB, and is localized in the cytoplasm.…”
