Effects of saffron and its constituents, crocin-1, crocin-2, and crocetin on α-synuclein fibrils

Abstract: Saffron, the stigma of Crocus sativus Linné (Iridaceae family), has been known to inhibit aggregation of β-amyloid, a nerve tissue protein. α-Synuclein (αS) is a 140-amino acid protein found abundantly in various regions of the brain. Its abnormal aggregation and accumulation in nerve tissue are said to cause neurodegenerative diseases such as Parkinson's disease, Lewy body dementia, and multiple-system atrophy. This study (part of this study was presented at the 137th Annual Meeting of the Pharmaceutical Soci… Show more

“…DLS and SEM results demonstrated that the size of all the produced CSNPs ranged from 150 to 500 nm, while their shape was spherical or irregular. Additionally, physicochemical characteristics of the prepared emulsions were enhanced, whereas their sunscreen protection factor was weak (SPF value from 2.15 to 4.85) [ 5 , 171 , 172 ].…”

“…Crocus sativus L., popularly known as saffron, is widely used for its color, flavor, and aroma in food and beverages. Saffron and its constituents, such as crocin-1, crocin-2, crocetin, safranal, and the crocetin structural analogs hexadecanedioic acid, norbixin, and trans-muconic acid, were found to affect α-syn fibrillation and aggregation (Inoue et al, 2018). However, some crocetin analogs failed to affect α-syn aggregation and dissociation.…”

“…Other studies report that water-soluble carotenoids such as crocins have antioxidant effects superior to α-tocopherol [67]. It was recently observed in a clinical study that high crocin I and crocin II contents (4000 and 1000 mg, respectively) inhibit β-amyloid and tau aggregation [70]. Apart from crocins, Asdaq and Inamdar [71] suggest that flavonols are responsible for the synergistic antihyperlipidemic and antioxidant potential of saffron.…”
