Effects of silencing cyclooxygenase-2 expression via RNA interference on the tumorigenicity of the SMMC-7721 human hepatocarcinoma cell line

Abstract: Abstract. We constructed a vector carrying a shRNA sequence against cyclooxygenase-2 (COX-2) that was subsequently transfected into the human hepatocarcinoma cell line SMMC-7721. Furthermore, we established a COX-2-deficient stable cell line and a model of tumor-shRNA transplantation in nude mice.

“…To our knowledge, there were few studies using lentivirus-mediated RNAi targeting COX-2 for determining the therapeutic significance of COX-2 in HCC. Liu et al, firstly explored the effects of silencing COX-2 expression via RNAi on the tumorigenicity of SMMC-7721 cell line 26. However, their study only provided primary results of the animal experiment rather than investigation of the detailed mechanism at the cellular level.…”

“…Hepatocellular carcinoma very often presents at an advanced stage and has a very poor prognosis [45]. GPS and five other kinds of traditional Chinese medicine extracts can lesion SMMC-7721 human hepatocellular carcinoma cells; however, the exact mechanism of action is not yet known [46]. 4.12.5.…”
