Effects of Some Metals on Carbonic Anhydrase from Brains of Rainbow Trout

Abstract: Carbonic anhydrase (CA) enzyme was purified from rainbow trout brain by Sepharose-4B-L: -tyrosine-sulfanilamide affinity chromatography. The enzyme was obtained with a specific activity of 2,275 EU mg(-1) and a yield of 22.5%. The sample obtained from the affinity column was used for kinetic properties and inhibition studies. Both optimum and stable pH were found as 9.0 in 1 M Tris-SO(4) at 4 degrees C, respectively. To check the purity and subunit molecular weight of enzyme, sodium dodecyl sulfate (SDS) polya… Show more

“…Soyut et al examined the inhibitory effects of cobalt, copper, zinc, silver and cadmium on carbonic anhydrase purified from brains of rainbow trout. They founded that cobalt, silver and cadmium inhibited the enzyme competitively, copper inhibited non-competitively, whereas zinc inhibited the enzyme uncompetitively 42 In another study, Soyut and Beydemir examined the effects of some metals (cobalt, copper, zinc, silver) on the CA activity of rainbow trout liver. Their results showed that cobalt and zinc inhibited the enzyme in a competitive manner and copper and silver inhibited the enzyme in an uncompetitive manner 43 .…”

“…Industrial waste is frequently emptied to the surroundings without any prerefining. As a result, in some areas, the natural habitat of animals and balance of nature are threatened by pollution in freshwaters, seas and atmosphere 18 . In the environment, heavy metal concentration is increased by quick industrialization, poor emission control, disordered urbanization, and risen motor traffic.…”
