Effects of Surgical Excision and Radiation on Medulloblastoma Cell Invasiveness

Abstract: In children, brain cancers can be particularly devastating; and medulloblastoma (MB), a primitive tumour of neuroectodermal origin, is among the most common. Rare in adults [1][2][3][4][5] , these tumours generally account for between 20 and 40% of paediatric intracranial malignancies worldwide [6][7][8][9][10][11][12] , and have an incidence of approximately 1-7 per million, depending upon the population studied and how medulloblastoma and the base populations are defined 11,[13][14][15][16] . They are especi… Show more

“…Medulloblastoma (MB) is a family of highly-invasive tumors most commonly diagnosed in the pediatric central nervous system (7, 28, 38, 41, 50) . While clinical treatments have more than doubled the overall 5-year survival rate to upwards of 60% (16, 28, 44, 52) , additional therapies are needed to target the aggressive MB migration that is uncharacteristic of other brain tumors, but is a hallmark of MB recurrence, metastasis, and radioresistivity (13, 21, 24, 28, 41, 55, 61, 70) .…”

“…These tumors are known to metastasize outside the central nervous system [3–6] and are likely to spread into bones, bone marrow, lymph nodes, liver, and lungs [7,8]. Compared with other childhood tumors, the median survival rate for medulloblastoma is poor, with a five-year survival rate of less than 50%.…”

“…DAOY cells were kindly provided by Dr Nada Jabado (Montreal Children's Hospital, McGill University Health Center, 2012), and have previously been characterized and validated . These cells were maintained in AMEM (alpha minimal essential medium; Wisent, St‐Bruno, QC, Canada) supplemented with 1% penicillin/streptomycin (Invitrogen, Burlington, ON, Canada) and 10% fetal bovine serum (Wisent).…”
