Effects of the pyrimidine-containing cytochrome P-450 inhibitor, fenarimol, on the formation of 20-OH ecdysone in flies

Abstract: The effect of fenarimol, a pyrimidine-containing cytochrome P-450 inhibitor, was tested in vitro on brain-ring gland complexes of Calliphora vicina (Dipt., Calliphoridae), and on microsomes prepared from the fat body of 0-h wandering stage larvae of Neobellieria bullata (Dipt., Sacrophagidae). Fenarimol had no influence on the formation of ecdysone, but it was an effective inhibitor of cytochrome P-450-dependent ecdysone 20-monooxygenase.

“…Some heterocyclic compounds containing pyridinyl, pyrimidinyl or imidazolyl groups and known as inhibitors of different cytochrome P450‐dependent monooxygenases also inhibit insect E‐20‐M, a P450‐dependent enzyme responsible for the activation of ecdysone to the moulting hormone 20E. Examples are metyrapone and PIM, as well as the fungicide fenarimol,18 a 14α‐demethylase inhibitor in fungal lanosterol biosynthesis. Metyrapone (though only at the high dose of 1000 mg kg −1 ) and PIM disturb the first moult of the fleshfly N bullata and the larvae die within the unshed, old cuticle.…”
