Effects of triethylene glycol dimethacrylate and hydroxyethyl methacrylate on macrophage polarization

Abstract: Aim To evaluate the effects of hydrophilic dental resin monomers, triethylene glycol dimethacrylate (TEGDMA) and hydroxyethyl methacrylate (HEMA), on the polarization of a human monocyte cell line (THP‐1). Methodology THP‐1 cells were treated with resin monomers at noncytotoxic concentrations for 48 h and were analysed for CD86 and CD206 expressions using flow cytometry. The cells were stimulated for polarization in the presence of resin monomers (co‐treatment) or after treatment with monomers (pre‐treatment).… Show more

“…The median fluorescence intensity of cells positive for the major histocompatibility complex class II molecule, HLA-DR, previously identified as an M1-like macrophage marker (42,43), were significantly upregulated in LPS and IFN-g treated macrophages when compared to M0 and IL-4-and IL-13-treated macrophages ( Figure 2E). The median fluorescence intensity of MR, previously used as an M2-like macrophage marker (42,44,45), was not increased ( Figure 2F). However, the percentage of cells positive for HLA-DR and MR, was also analysed ( Figure 2G) and HLA-DR was significantly upregulated in LPS and IFNg treated macrophages ( Figure 2H) and MR was significantly upregulated in IL-4-and IL-13-treated macrophages ( Figure 2I).…”

“…M2 phenotype of macrophages has been proved to promote cutaneous wound healing ( Zhang et al, 2010 ). Therefore, we next analyzed the M2 macrophage markers ARG1 and CD206 to explore the distribution of macrophages at the site of the icariin+PEG hydrogel treatment ( Kim et al, 2019 ). As shown in Figures 4A,B , the PEG hydrogel group had fewer ARG1-positive cells; in contrast, their distribution in the icariin+PEG hydrogel group was more widespread under the skin of the wound.…”

“…Statistically, elongated macrophages treated with AAB scaffold had higher ratio of CD206/DAPI (Figure 4D ), indicating the polarization state of macrophage is related to M2 phenotype. [ 67 ] Western blot and qRT‐PCR analysis were also applied to further confirm the effects of AAB on macrophage polarization (Figure 4F ). After incubation with AAB for 24 h, the expression of M2 macrophage‐related protein, including CD206 and Arg‐1, were increased compared to control group (Figure 4 G,H).…”
