This study aimed to evaluate the antimicrobial activity of a dentifrice containing an alcoholic extract of rosemary on oral bacteria, compared to a commercially available herbal dentifrice. Standard strains of Streptococcus mutans (ATCC 25175), Streptococcus oralis (ATCC 9811) and Lactobacillus rhamnosus (ATCC 7469) were used, as well as different toothpastes based on rosemary (TR), on propolis (TH), triclosan (positive control) (TPC) and non-fluoridated dentifrice (negative control) (TNC). Bacteria were seeded in Petri dishes and paper discs soaked with dilutions of dentifrice placed on the plates. The inhibition halos were analyzed. It was observed that TR did not show statistical difference in relation to the TH to inhibit S. mutans and S. oralis, while TH was more active against L. rhamnosus. The toothpaste containing rosemary extract had the ability to inhibit the growth of S. mutans, S. oralis and L. rhamnosus, revealing an antimicrobial activity similar to commercially available toothpastes for inhibition of S. mutans and S. oralis.

The objective of this study was to evaluate the effects of Cymbopogon citratus essential oil and its main compound (citral) against primary dental colonizers and caries-related species. Chemical characterization of the essential oil was performed by gas chromatography/mass spectroscopy (GC/MS), and the main compound was determined. Antimicrobial activity was tested against Actinomyces naeslundii, Lactobacillus acidophilus, S. gordonii, S. mitis, S. mutans, S. sanguinis and S. sobrinus. Minimum inhibitory and bactericide concentrations were determined by broth microdilution assay for streptococci and lactobacilli reference, and for clinical strains. The effect of the essential oil on bacterial adhesion and biofilm formation/disruption was investigated. Negative (without treatment) and positive controls (chlorhexidine) were used. The effect of citral on preformed biofilm was also tested using the same methodology. Monospecies and microcosm biofilms were tested. ANOVA or Kruskal-Wallis tests were used (α=0.05). Cytotoxicity of the essential oil to human keratinocytes was performed by MTT assay. GC/MS demonstrated one major component (citral). The essential oil showed an inhibitory effect on all tested bacterial species, including S. mutans and L. acidophilus. Essential oil of C. citratus (10X MIC) reduced the number of viable cells of lactobacilli and streptococci biofilms (p < 0.05). The essential oil inhibited adhesion of caries-related polymicrobial biofilm to dental enamel (p < 0.01). Citral significantly reduced the number of viable cells of streptococci biofilm (p < 0.001). The essential oil showed low cytotoxicity to human keratinocytes. Based on these findings, this study can contribute to the development of new formulations for products like mouthwash, against dental biofilms.

ResumoIntrodução: A avaliação microbiológica em produtos de higiene pessoal constitui uma etapa importante no que se refere à segurança do usuário e à qualidade do produto, visto que a carga microbiana elevada pode acarretar problemas de saúde, especialmente em pessoas imunocomprometidas. Objetivo: Verificar o cumprimento das exigências acerca da qualidade microbiológica de cremes e géis dentais adquiridos comercialmente. Material e método: Realizou-se a contagem de bactérias e fungos viáveis totais e pesquisa dos patógenos E. coli, Salmonella sp., S. aureus e P. aeruginosa em 21 amostras. Resultado: Das amostras analisadas, 52,0% apresentaram crescimento microbiano e 28,6% e 0,21% apresentaram contaminação fúngica e bacteriana, respectivamente, acima dos limites descritos na Farmacopeia Brasileira para preparações de uso tópico (máximo permitido 2 × 10 2 UFC/g de bactérias e 2 × 10 1 UFC/g de fungos). Nenhuma amostra apresentou os patógenos pesquisados E. coli, Salmonella sp., S. aureus e P. aeruginosa. Conclusão: Estes resultados indicam que muitos produtos disponíveis no mercado apresentam qualidade inadequada, demonstrando falhas no controle de qualidade. Para prevenir esta situação, faz-se necessária fiscalização rigorosa e adoção de medidas regulamentadoras e educacionais aliadas ao seguimento das Boas Práticas de Fabricação pelas indústrias fabricantes.Descritores: Contaminação microbiológica; controle de qualidade; cremes dentais; géis dentais. AbstractIntroduction: Microbiological evaluation in personal care products is important to guarantee user safety and product quality since microbial contamination elicits health problems especially in immunocompromised patients. Objective: To verify the compliance with the requirements regarding the microbiological quality of creams and dental gels acquired commercially. Material and method: Microbiological contamination was performed through total bacterial and fungal viable count and research of E. coli, Salmonella sp., S. aureus and P. aeruginosa in twenty-one samples. Result: 52.0% of sample presented microbial growth, and 28.6% and 0.21% presented fungal and bacterial contamination, respectively, exceeding the limits described in Brazilian Pharmacopoeia for topical preparations (maximum allowable 2 × 10 2 CFU / g of bacteria and 2 × 10 1 CFU / g yeast). None of the researched pathogens E. coli, Salmonella sp., S. aureus and P. aeruginosa were found. Conclusion: These results indicate that many products available in the market present inadequate quality, demonstrating quality control failures. Rigorous inspection and adoption of regulatory and educational measures aligned with the compliance of Good Manufacturing Practices by manufactures are needed to prevent this situation.

Abstract: Supernatants based on herbal- and chitosan-based toothpastes have comparable immediate and ongoing antibacterial efficacies as chlorhexidine. Natural antimicrobials and chlorhexidine absorb in oral biofilms which contributes to their substantive action.
