This study aimed to explore different substances (or cold sea water) as potential anesthetic agents to facilitate short-term handling in Octopus maya juveniles. We investigated oxygen consumption before (baseline), during (first 600 s of exposure) and after anesthesia (recovery) of octopuses ( n = 98; 1.67 ± 0.5 g) exposed to cold sea water (SW; 11 and 13°C), ethanol (EtOH; 0.5; 1.5 and 3.0%), magnesium chloride (MgCl 2 ; 0.75; 1.5 and 3.75%), ethanol combined with magnesium chloride (Mix; 1.5:0.75%; 0.75:1.13%; and 2.25:0.37%) and clove oil (0.15 mL L –1 ). After exposure, the animals were handled for 180 s (exposed to air) and weighted. Two experimental groups not exposed to anesthetics (with or without handling) were also evaluated. The criteria for general anesthesia were analysed. Times of induction and recovery, incidence of attack response after recovery and possible longer-term effects of repeated general anesthesia on growth and mortality of the octopuses were evaluated. During anesthesia, O. maya juveniles exposed to SW (11 and 13°C), EtOH (0.5; 1.5 and 3.0%), Mix (0.75:1.13%), and clove oil, presented a significant decrease on oxygen consumption. In animals exposed to different concentrations of EtOH and Mix 0.75:1.13%, this decrease was registered after an increase on oxygen consumption. Animals exposed to MgCl 2 did not show significant changes on oxygen consumption, except for animals exposed MgCl 2 3.75%, which showed a significant increase on oxygen consumption. At the end of recovery, except for octopuses exposed to clove oil and MgCl 2 0.75%, the values of oxygen consumption observed were comparable to the ones registered during baseline. Animals exposed to SW 11°C, EtOH 3.0%, Mix 1.5:0.75% and MgCl 2 3.75% fulfilled the criteria defined for general anesthesia. Exposure to MgCl 2 (all concentrations), SW 13°C and clove oil reduced or inhibited the incidence of attack response after recovery. Except for animals exposed to clove oil, growth of the juveniles was not affected by the exposure to the different substances. Short-term handling (180 s) of O. maya juveniles can eventually be carried out without anesthesia. However, to facilitated handling, we suggest the use of EtOH 3.0% or cold sea water 11°C.

Abstract:The Indian clove, Syzygium aromaticum [L] Merr. et Perry, has been popularly used in the treatment of several diseases. A more recent use of this spice in Brazil is a homemade formulation, based on the ethanolic extraction of the dried flower buds of S. aromaticum, which has shown to be efficient as mosquitoes repellent. Here we evaluated, through the development of a GC analytical technique, the concentrations of the main components of the ethanolic extract of Indian cloves (eugenol and eugenyl acetate), and possible responsible for the repellent activity, in this homemade formulation. Our results corroborated the efficiency of this protocol to extract eugenol and eugenyl acetate and established the ideal conditions for extraction.Keywords: Eugenol; quantification; eugenyl acetate; repellent; indian cloves. ResumoO cravo-da-índia, Syzygium aromaticum [L] Merr. Perry et, é popularmente usado no tratamento de várias doenças. Uma utilização mais recente dessa especiaria no Brasil é uma formulação caseira, com base na extração de etanol dos botões florais secos de S. aromaticum, que se mostrou eficiente como repelente contra mosquitos. Aqui nós avaliamos por meio do desenvolvimento de uma técnica analítica utilizando CG, as concentrações dos principais componentes do extrato etanólico do cravo (eugenol e acetato de eugenila), possíveis responsáveis pela atividade repelente, nesta formulação caseira. Nossos resultados corroboram a eficiência deste protocolo para extrair eugenol e acetato de eugenila, e estabeleceu as condições ideais para extração, além da caracterização dos compostos.Palavras-chave: Eugenol; quantificação; acetato de eugenila; repelentes; cravo da Índia.

“…O óleo de cravo tem a vantagem de não irritar a mucosa e possuir maior facilidade de decomposição no meio ambiente. 53 Ensaios para atividade anti-inflamatória através do método de edema de pata mostraram que a administração oral do eugenol (400 mg/Kg) em ratos inibiu significativamente o edema, sendo a taxa de inibição comparável a dos anti-inflamatórios não esteroidais (AINES), tais como a indometacina e o celecoxibe. Houve também uma redução na formação de exsudato pleural e desta forma, o eugenol apresentou efeito farmacológico semelhante aos antagonistas das enzimas COX-1 e COX-2.…”

Abstract:The Indian clove (Syzygium aromaticum, Myrtaceae) is a well appreciated spice since the ancient times not only by its flavor and culinary qualities, but also because of its therapeutic uses. Several popular and traditional applications have been reported in literature as well as numerous scientific studies on its activities. In this work we present a synopsis and discussion on S. aromaticum to provide the readers some useful information on the historical aspects, the chemical composition and its large variety of potential applications for the treatment of a number of illnesses.Keywords: Syzygium aromaticum; essential oil of Indian cloves; eugenol. ResumoO cravo-da-índia (Syzygium aromaticum, Myrtaceae) é uma especiaria muito apreciada desde a antiguidade, não só por seu sabor e qualidades culinárias, mas também por suas utilizações terapêuticas. Várias aplicações desta especiaria na cultura popular têm sido relatadas na literatura bem como diversos estudos científicos sobre esses usos. Neste trabalho, apresentamos uma sinopse e discussão dos relatos da literatura nos últimos 20 anos (tendo o SCOPUS como principal base de dados) sobre o óleo essencial do S. aromaticum, com o objetivo de proporcionar aos leitores algumas informações úteis sobre os aspectos históricos, a composição química e a grande variedade de suas aplicações potenciais no tratamento de uma série de doenças.Palavras-chave: Syzygium aromaticum; óleo de cravo; eugenol.
