Efficient Access to Nonhydrolyzable Initiator tRNA Based on the Synthesis of 3′‐Azido‐3′‐Deoxyadenosine RNA

Abstract: Flexibility exercised: Hydrolysis‐resistant 3′‐aminoacyl‐tRNA conjugates that contain a stable amide linkage instead of the natural ester are valuable substrates for biochemical studies of ribosomal processes. In a novel preparation of the stable E. coli initiator tRNA derivative 3′‐(N‐formylmethionyl)amino‐tRNAfMet the key feature is the synthesis of 3′‐azido oligoribonucleotides using a new functionalized solid support.

“…As III exhibits a higher stability towards oxidation than P III in phosphine-derived ligands. We therefore assumed that Staudinger-type reactions between phosphines and azides, [5,18] which prevent the direct Table 1. Identification of a suitable catalyst system for the Stille-Migita reaction on the unprotected 5-I-(d)UpG dinucleotide.…”

“…4D). 31 At reaction conditions reported in the literature, we observed decomposition of the azide to an amine, very likely owing to the known acid and light sensitivity of azides. 32 Therefore, the original protocol was slightly changed; reactions were performed in the dark with strict control of the pH during all synthesis and purification steps, thus preventing azide decomposition.…”
