Efficient gene delivery of primary human cells using peptide linked polyethylenimine polymer hybrid

Abstract: Polyethylenimine (PEI) based polymers are efficient agents for cell transfection. However, their use has been hampered due to high cell death associated with transfection thereby resulting in low efficiency of gene delivery within the cells. To circumvent the problem of cellular toxicity, metal binding peptides were linked to PEI. Eight peptide-PEI derivatives were synthesized to improve cell survival and transfection efficiency. TAT linked PEI was used as a control polymer. Peptides linked with PEI amines for… Show more

“…To investigate if our carrier system provided physical protection of the complexed DNA against intracellular DNases, a stabilization assay was used . DNases are ubiquitously present in cells mostly in the nucleus and take part in many cellular activities concerned with DNA processing such as DNA repair and apoptosis .…”

“…On the other hand, upon crosslinking PEI with GDE, the most distinct peaks are 1051 and 1109 cm −1 coming from alcohol groups, for O‐H bending and C‐O stretching from secondary alcohol, respectively. PEI cryogels were also treated with 200 mL of 1.0 M NaOH, and 0.1 M HCl for 4 h assess the generated peaks as given in Figures (a) and , respectively, and the corresponding peaks are shown in the figure. The quarternized amine peaks are clearly seen at 1660 cm −1 for all the PEI materials.…”

“…Guanidinylation or addition of α-tocopherol succinate enhanced the cellular uptake of chitosan complexes [46,47], whereas stearoyl groups facilitate cell penetration of polypeptides [48]. Some peptides have a natural cell penetration capability which can be linked to other molecular structures [49].…”
