Efficient screening of pancreatic lipase inhibitors from cod meat hydrolysate through ligand fishing strategy

Abstract: Obesity has become an increasingly serious public health problem. Pancreatic lipase (PL) is identified as a ideal target for the prevention and treatment of obesity. Orlistat, the only approved PL inhibitor (PLI), is a powerful weight loss drug but has many side effects. Therefore, there is an urgent need to discover powerful PLIs with high safety. Protein hydrolysate has been demonstrated to be a treasure trove of PLIs, but recognizing responsible functional peptides from them is like looking for a needle in … Show more

“…The PL, also known as triacylglycerol esterase, is one of the most essential lipases for the intestinal absorption of fatty acids. In human PL, Ser-152, His-263, and Asp-176 make up a typical catalytic triad, which is highly conserved [ 34 ]. The problem of obesity can be controlled by inhibition of PL activity.…”

“…Two pancreatic lipase inhibitory peptides were isolated from cod meat hydrolysate, namely GSPPPSG and KLEG-DLK, with IC 50 values of 0.60 and 1.08 mg/mL (Tian et al 2022). Fish goby protein hydrolysates fed to high fat and fructose diet-fed rats showed good efficacy in lowering serum TC, TG, and LDL-C levels as well as hepatic Ma et al Food Production, Processing and Nutrition (2023) 5:10 TC and TG contents, whilst increasing the serum HDL-C content (Nasri et al 2018).…”
