Egg-to-Embryo Transition Is Driven by Differential Responses to Ca2+ Oscillation Number

Abstract: Ca(2+) oscillations and signaling represent a basic mechanism for controlling many cellular events. Activation of mouse eggs entrains a temporal series of Ca(2+)-dependent events that include cortical granule exocytosis, cell cycle resumption with concomitant decreases in MPF and MAP kinase activities, and recruitment of maternal mRNAs. The outcome is a switch in cellular differentiation, i.e., the conversion of the egg into the zygote. By activating mouse eggs with experimentally controlled and precisely defi… Show more

“…Subsequent IP 3 binding to its own receptor on the endoplasmic reticulum (ER) [19] opens Ca 2+ channels, which release Ca 2+ from the ER into the cytoplasm of the oocyte [19]. The resulting Ca 2+ oscillations within the oocyte [18,19] disseminate from the cortical region throughout the ooplasm [20,21] at 10-to 20-min intervals [22], triggering oocyte activation [23] to initiate oocyte fertilization and preimplantation embryogenesis [8,14,17].…”

“…In mouse, inhibiting Ca 2ϩ release at fertilization prevents many activationassociated changes in protein synthesis (Xu et al, 1994). Reciprocally, translation can be stimulated by artificially increasing Ca 2ϩ within the unfertilized egg (Ducibella et al, 2002). A hypothesis for the molecular mechanism of Ca 2ϩ -induced translation in mouse has been inferred from another murine cell type, hippocampal neurons .…”

“…Moreover, the calcium oscillations triggered by the sperm cells function not only in inducing 70 resumption of meiosis but also in many other events [26]. For example, recruitment of specific maternal RNAs [27,28] which is essential for activation of zygotic genome [29] and may be extended to other unknown functions. In addition, using bovine oocytes harvested from ovaries stored for 26-30 h resulted in the great reduction of the blastocyst yield after ICSI [30], though the developmental potential of these oocytes after in vitro fertilizaton (IVF) or somatic cell nuclear 75 transplantation was not impaired [30,31].…”
