Eicosanoid formation in the rat cerebral cortex

Abstract: Despite the extensive literature on brain eicosanoids, no information is available on the cellular source of individual compounds in the mature organ and the relative contribution of different cell types to the total synthetic product. To address this problem, neurons and glia were isolated from the cerebral cortex of the adult rat by a process comprising, in order, trypsinization, selective sieving, differential centrifugation, and density gradient centrifugation. Enrichment of cells in the appropriate fracti… Show more

“…PGE 2 and PGD 2 are the predominant PG species in the brain, and PGE 2 is most widely used as a marker of COX activity in neuronsFspecially in the cortex (Bishai and Coceani, 1992). Samples were purified using Amprept (RPN1900 Amersham, Buckinghamshire, UK) minicolums.…”

“…Prostaglandin E 2 (PGE 2 ) is the most abundant prostaglandin in the brain (Bishai and Coceani, 1992) and is considered to play an essential role in several pathological states. In the central nervous system (CNS), PGE 2 is responsible for fever induction (Ushikubi et al, 1998) and participates in eliciting pain hypersensitivity (Samad et al, 2001).…”

“…PGE 2 is a major prostanoid that is involved in a myriad CNS functions (Wolfe and Coceani, 1979;Ho et al 1999;McCullough et al 2004). PGE 2 is synthesized in the CNS by neurons and glia cells (Bishai and Coceani, 1992;Guay et al 2004). Cyclooxygenase 2 (COX-2) converts free arachidonic acid to the intermediate prostaglandin H 2 (PGH 2 ).…”
