Electrochemical Imaging of Dopamine Release from Three-Dimensional-Cultured PC12 Cells Using Large-Scale Integration-Based Amperometric Sensors

Abstract: In the present study, we used a large-scale integration (LSI)-based amperometric sensor array system, designated Bio-LSI, to image dopamine release from three-dimensional (3D)-cultured PC12 cells (PC12 spheroids). The Bio-LSI device consists of 400 sensor electrodes with a pitch of 250 μm for rapid electrochemical imaging of large areas. PC12 spheroids were stimulated with K(+) to release dopamine. Poststimulation dopamine release from the PC12 spheroids was electrochemically imaged using the Bio-LSI device. B… Show more

“…As an imaging tool, the size of the device, and inherently needed electrode separation distance, will limit its use. Due to the limitations of device size and structure, this type of imaging will typically have lower resolution [134]. Matsue and coworkers [135] used an amperometric sensor array device at a size-scale suited to cell clusters.…”

“…CV is performed by cycling the potential between the working electrode and the reference electrode, and measuring the resulting current. This technique is widely used in the development of cost-effective, simple and rapid methods for Ab detection [12]. There are several research studies that used CV as their main tool and have obtained great results.…”

“…By coating the sensor surface with gel polymer containing horseradish peroxidase, the production and diffusion of H 2 O 2 catalyzed by glucose oxidase was clearly visualized. (9) Reported applications of the BioLSI includes the characterization of an embryoid body by monitoring the activity of alkaline phosphatase, (10) the visualization of dopamine release from spheroid cells, (11) and conductivity imaging.
