Electrochemical measurement of antibody-antigen recognition biophysics: Thermodynamics and kinetics of human chorionic gonadotropin (hCG) binding to redox-tagged antibodies

Abstract: The thermodynamics and kinetics of antigen binding under diffusive conditions to an electrode surface modified with ferrocene--tagged antibodies is studied within this work, and realised experimentally for the case of human chorionic gonadotropin (hGC) as the antigen with monoclonal anti--hCG antibodies immobilised on an electrode surface via a molecular wire platform. The formation of the antigen--antibody complex is monitored through the blocking of the ferrocene voltammetry, thereby enabling the fractional … Show more

“…[26] linear range of 0.001-0.2 IU/mL with a LOD of 0.6 mIU/mL. The authors note that IU (international unit)/mL refers to the international arbitrary amount of a substance agreed upon by scientists and doctors; in the case of hCG, a reading of above 25 mIU/mL is considered a positive result for pregnancy [109], and a result <5 mIU/mL generally indicates a negative test [110]. Another interesting use of AuNPs was reported by Li et al [26] who created a hybrid array biosensor for the detection of penicillin and tetracycline using penicillinase and L-cysteine as biorecognition elements, respectively (Fig.…”

“…A decrease of signal intensity of the oxidation of ferrocene label when incubated with specific antigen was observed. Very recently, the thermodynamics and kinetics of human chorionic gonadotropin (hCG) binding to the surface of electrodes modified with ferrocene-tagged antibodies theoretically and experimentally have been studied [54]. Upon the formation of antigen-antibody complex, the blocking of ferrocene voltammetry occurs.…”
