Electron cryotomography sample preparation using the Vitrobot

Abstract: Electron cryotomography is the highest-resolution structural technique currently available that can be applied to unique objects such as flexible large protein complexes, irregular viruses, organelles and small cells. Specimens are preserved in a near-native, 'frozen-hydrated' state by vitrification. The thickness of the vitreous ice must be optimized for each specimen, and gold fiducials are typically added to facilitate image alignment. Here, we describe in detail our protocols for electron cryotomography sa… Show more

“…(36) and T. maritima cells were treated with 1 mg/mL polymyxin B for 10 h on ice. Cultures were plunge-frozen across EM grids as described (37). Standard EM tilt series were collected on 300 kV electron cryo-microscopes and 3D reconstructions were calculated as described (38,39).…”

“…Other plungers that can be used and the protocols to use them are the Vitrobot (http://www.fei.com/) (see ref. 23), the Gatan cryoplunge (http://www.gatan.com/) and a time-resolved plunge-freezing apparatus. The time-resolved unit, which is made to order, is useful when attempting to resolve reactions that occur in the 10-ms timescale24.…”

“…The cryo-samples were prepared as reported previously (34). Briefly, 3 l of isolated axonemes (labeled or unlabeled for control) in HMEEK buffer and 1 l of 5-fold concentrated and BSA-treated 10 nm colloidal gold (Sigma) (35) were applied to Quantifoil holey carbon grids (copper R2/2, Quantifoil Micro Tools GmbH, Jena, Germany) that were previously coated with 10 nm colloidal gold. Excess fluid was blotted from the grid for ϳ2 s with Whatman filter paper and then plunge frozen in liquid ethane using a homemade plunge freezer to achieve sample vitrification.…”
