The herpes simplex virus type 1 temperature-sensitive (ts) DNA-positive mutant ts1203 has been characterized. The ts lesion in ts1203 was located by marker rescue within the coding region of gene UL28. Nuclei of cells infected with ts1203 at the non-permissive temperature (NPT) contained large numbers ofcapsids with a uniform morphology. These capsids lacked DNA but had a defined internal structure. No full capsids were detected at the NPT, suggesting that tsl203 was unable to package viral DNA. In this respect tsl203 is similar to ts1201 which has a defect in gene UL26. The capsids made by ts1203 at the NPT, however, contained a more compact internal structure than those of tsl201. In addition, ts1203 capsids were dispersed throughout the nucleus whereas tsl201 capsids were frequently found clustered together in large arrays. Southern blot and sedimentation analyses of viral DNA confirmed that ts1203 had an encapsidation defect and showed that most of the mutant DNA at the NPT was of a high Mr. The effect of the ts1203 mutation could not be reversed in the absence of de novo protein synthesis by transferring mutant-infected cells from the NPT to the permissive temperature.

SUMMARYWe report the ultrastructural features associated with wild-type and temperaturesensitive (ts) mutant infection at non-permissive temperature for each of 18 herpes simplex virus type 1 (HSV-1) (strain 17) ts mutants. The mutants were classified by their ability to induce nucleocapsid-related structures : class I failed to induce any, class II induced empty and partially filled structures while class III induced all of the identified structures including those containing an electron-dense core. The time when expression of the ts lesion blocked virus replication was estimated for most mutants; this allows both mutant gene expression and the resulting ultrastructural features to be correlated with the sequence of virus replicative events. Three ts mutant-triggered features not previously described in HSV-l-infected cells were also recognized: a modification of rough endoplasmic reticulum, intranuclear accumulation of enveloped virus particles and cytoplasmic accumulation of novel doughnut-shaped particles having a concentric double ring appearance in thin section.

Veligers removed from brooding T~ostrea chilens~s (Philippi, 1845) experienced -95% mortalities over 3 to 4 d at 16 to 18°C that appeared to be associated with a herpes-like virus. Ultrastructural observations of post-removal veligers showed the presence of early viral replication or putative latent stages at 4 h, all stages of replication at low levels at 24 h, which increased to high levels at 48 h, followed by mortalities at 72 h onwards. Initially, infected interstitial or epithelia1 cells had an enlarged nucleus with a wavy outline in which heterochromatin was marginated. With continued increase in size, nuclei became smooth in outline with reduction or loss of heterochromatin. Capsids with lucent cores (LCC) and empty capsids appeared in the nucleus, often in association with tubular structures -65 nm in diameter that were composed of subunits in a helical configuration that contained a tubular core -35 nm in diameter. Empty capsids and LCC sometimes occurred in paracrystalline arrays. Partial nucleolar disaggregation and encapsidation of dense fibrillar material preceded envelopment entering and de-envelopment leaving the perinuclear cisterna, tegumentation in cytoplasmic vesicles, and egress. Groups of dense cytoplasmic filaments 30 to 35 nm in diameter occurred in some infected cells. Apparently normal cells with a few intranuclear empty capsids and/or LCC at 4 h postremoval may represent latent infections. Replication was not observed in larvae held at 24 to 27'C. but a few cells had enlarged hypochromatic karyolytic nuclei, and 1 to 2 capsids were observed in them, at 48 h. Viral replication was similar to that of ranid herpesvirus 1 (Lucke tumour) infections. This is the fifth ostreid species from which herpesviruses have been reported.
